双酚A双(二苯基磷酸酯)(BDP)是国内需求高的一种低聚有机磷酸酯阻燃剂,其在太湖沉积物中的赋存与单体有机磷酸酯的总量相当,鉴于其代谢产物双酚A的危害,BDP在水生植物根际的降解、以及被植物吸收并在体内代谢又经腐解释放的行为亟待解析。本项目拟针对生态修复常用的挺水植物,研究BDP在其根际和体内的迁移转化机制。首先,研究根际细菌和根系分泌物的响应,用根系分泌物培养优势菌群并筛选降解菌,阐明根系分泌物介导下根际优势菌群降解BDP的调控机制;其次,研究植物吸收BDP的动力学及BDP在皮层、维管束和亚细胞的分布,结合蛋白组学分析揭示植物吸收和运输BDP的途径及转运蛋白;最后,采用转录组学技术分析植物体内BDP代谢酶,并研究微生物对腐解释放的作用,探明植物体内BDP的酶代谢和腐解释放过程。本项目有助于预测真实环境中BDP的归趋,支持BDP生态风险评估模型的建立,推动BDP污染治理的植物修复技术应用。

Bisphenol A bis(diphenyl phosphate) (BDP) is a popular oligomeric organophosphorus flame retardant with high demand in China. It has been detected in the sediment of Taihu lake with a concentration comparable to the sum of traditional monomeric organophosphorus flame retardants. Due to its harmful metabolite bisphenol A, it should be paid great attention to the biodegradation of BDP in the rhizosphere of aquatic plants, and also to the uptake of BDP by aquatic plants followed by the in vivo transformation and the decomposing release. The main objective of this project is to explore the rhizospheric and in vivo translocation and transformation mechanisms of BDP for the emergent macrophytes commonly used in ecological restoration. Firstly, we will investigate the characteristics of rhizosphere bacteria and root exudates in response to BDP, cultivate the dominant bacteria with root exudates, and screen the BDP degrading bacteria to clarify how root exudates regulate the dominant rhizosphere bacteria to degrade BDP. Secondly, we will study the uptake kinetics of BDP and the distribution of BDP in the cortex, vascular bundles and subcelluar fraction. And then combined with the proteomic analysis, we will reveal the uptake and transportation pathway of BDP by plants and the associated transport proteins. Finally, transcriptomics will be used to analyze the enzymes able to metabolize BDP in plants, and the role of microorganisms in plant decomposition will be also studied, aiming to elucidate the enzyme-catalyzed metabolism of BDP inside plants and the release of BDP and its metabolites via decomposing process. This project will help predict the fate of BDP in the real environment, support the establishment of ecological risk assessment model for BDP, and promote the application of phytoremediation technology for BDP pollution control.
双酚A双(二苯基磷酸酯)(BDP)是国内需求高的一种低聚有机磷酸酯阻燃剂,其代谢产物双酚A(BPA)潜在危害大,亟需了解BDP在环境介质中的赋存特征、挺水植物吸收运输BDP的潜力、BDP在根际的降解行为、以及植物体内BDP的酶代谢和腐解释放过程。本项目首先研究了转运蛋白协助下挺水植物菖蒲和再力花吸收运输BDP的途径,发现两种植物的根部都可以通过质外体、共质体和跨膜途径吸收BDP,兼有主动运输和被动运输。菖蒲倾向于在蒸腾流驱动下向上运输BDP,而再力花具有在地上部类皮层组织通过质外体途径运输BDP的潜力。潜在的BDP转运蛋白包括ABC转运蛋白、水通道蛋白、阴离子通道蛋白和脂质转运蛋白(如非特异性脂质转运蛋白)。鉴于再力花吸收BDP的潜力比菖蒲更高,进一步研究了根系分泌物调控下BDP在再力花根际的降解行为,发现在BDP胁迫下再力花的根系分泌物和根际微生物群落皆发生了明显改变。可以有效促进BDP降解的根系分泌物包括有机酸(奎尼酸、苹果酸、琥珀酸、乳酸、柠檬酸和酒石酸)和酚酸(原儿茶酸、对羟基苯甲酸、羟基肉桂酸、阿魏酸),具备降解BDP潜力的根际细菌有氢噬胞菌属和草酸杆菌属,BDP经根际细菌降解可以生成BPA和磷酸二苯酯(DPHP)。通过继续研究再力花体内BDP的酶代谢和腐解释放过程,发现BDP在植物体内通过酶代谢也可以生成BPA和DPHP,发挥作用的酶包括细胞色素P450酶、谷胱甘肽转移酶、羧酸酯酶和酸性磷酸酶。植物组织经腐败分解后,体内的BDP、BPA和DPHP可以重新释放到水体中。在上述研究开展的同时实地调研了环境介质中有机磷酸酯(OPEs)的赋存,发现太湖水体和沉积物中不同类型OPEs的赋存浓度排序为:氯代烷基OPEs>烷基OPEs>芳香基OPEs≈低聚OPEs,水体中的OPEs整体上呈现中等生态风险。另外,电缆园附近农田土壤中存在OPEs和邻苯二甲酸酯(PAEs)的复合污染,其中BDP较易被土壤有机碳吸附,OPEs和PAEs的混合风险熵在土壤表层为高等。项目研究结果既可以为BDP污染控制的植物修复技术提供思路,也可以为BDP的生态风险评估提供数据支持。
