Guiding bone regeneration (GBR) is an effective method for curing the bone defects. There is still no GBR materials in clinical practice, which could meet all the following demands including in vivo biodegradation, high strength and rigidity (high Young’s modulus) (ensuring the structure stability of the bone defects area). In this project, we plan to use the biodegradable chitin and hydroxylapatite (HA) for constructing the high strength and stiff composite films. Considering the negative effects of the two factors (1. the weak chitin/HA interaction, and 2. the small mobility of the chitin chains resulting from the hydrogen bonding interaction mode among the chitin chains) on the mechanical properties of the composite materials, the noncovalent receptor will be introduced to enhance the chitin/HA interface interaction and the chitin network through forming the noncovalent bonds at the chitin/HA interface and within the chitin networks. Moreover, the biomaterial attributes of chitin and HA will be maintained in this process because that no new covalent bond will form. The further nacre structure design and hot press induced self-assembly method will be applied to produce the chitin/HA composite materials with the hierarchical structure, which exhibits the high strength and rigidity at both the tensile and bending state. The mesenchymal stem cell and the calvarial defect model will be used to evaluate the affections and action mechanism of the chitin/HA composite materials structure and properties on the bone defects regeneration. It is expected that the project will open a new avenue for enhancing the chitin network and chitin based hybrid material interface interaction and providing new method and strategy for GBR materials preparation, showing important scientific significance and practical value.
