英文摘要
Familial exudative vitreoretinopathy (FEVR) is an ophthalmic disease caused by disordered retinal vascular development. FEVR has genetic and clinical heterogeneity and lacks treatment methods for it. Previous studies have shown that Norrin/β-catenin signaling pathway-related gene mutations that reduce the activity of this pathway are considered to be the main cause of FEVR, while we found that mutations lead to hyperactivation of Norrin/ β-catenin pathway can also cause FEVR, but the specific pathogenic molecular mechanism remains to be elucidated. This project intends to use the two groups of FEVR mouse models that have been previously constructed due to abnormal hyperactivation and hypoactivation of the Norrin/β-catenin pathway, and carry out single-cell transcriptome and proteomic analysis to find the downstream genes and cascade signals. Then we will conduct immunoblot, immunohistochemistry, and AAV virus-mediated rescue experiments to verify the proposed pathway and finally elucidated the pathogenic mechanism of these two types of mutations, and reveal the signal regulation processes related to clinical heterogeneity of FEVR. This project is expected to provide new targets for the diagnosis and treatment of FEVR, which has basic and clinical significance.
家族性渗出性玻璃体视网膜病变(Familial exudative vitreoretinopathy, FEVR)是一种视网膜血管发育紊乱导致的眼科疾病,具有遗传和临床异质性,且对其治疗手段匮乏。以往研究表明,Norrin/β-catenin通路相关基因突变致该通路失活是导致FEVR的主要原因,但我们发现Norrin/β-catenin通路异常激活亦可导致FEVR,但具体致病机制有待阐明。在国家自然科学基金项目支持下,我们原创性地构建了活性升高和降低的两组 FEVR 小鼠模型,对Norrin/β-catenin 信号调控网络与 FEVR 发生、发展之间的关系进行了深入系统的研究,探讨了不同类型突变之间致病机制的异同,应用转录组、蛋白组、单细胞分析,结合细胞生物学、生物化学和病理学数据,我们发现TWIST1通过增强Norrin/β-catenin通路,并通过视网膜胶质细胞增生促进VEGFA分泌,上调一组FEVR相关基因的表达,从而促进FEVR进程。同时我们阐明了FEVR中视网膜内皮细胞的表达谱,并确定了 4 个潜在的关键靶点(Ctss, Ccl4, Ccl3和Apoe)。本项目揭示了基于不同类型突变调控Norrin/β-catenin 信号通路引起的FEVR的新的分子机制与临床表型异质性。明确了Norrin/β-catenin通路下游关键基因和级联信号。为以病理新生血管形成为特征的疾病的药物设计和基因治疗研究提供了新的见解。
