“多基原”中药的品质和生物效应等同性问题是中药现代化进程中普遍关注的难题,而仅仅阐述化学成分的差异性难以全面的揭示品种之间的差异化的科学内涵。中国药典中车前子来源于两个基原,课题组研究发现这两个品种在化学成分上和治疗糖尿病时均有显著性的差异,但对其引发的科学内涵尚不明确。基于此,本研究在中医理论指导下,以车前子的两个基原品种的为研究对象,以防治糖尿病为方向,采用中药化学、分析化学、中药药理学和药物代谢组学相结合的研究方法和技术,采用化学/药效关联分析,鉴定药效成分(群)的差异性问题,阐述多基原车前子的药效物质基础及其在体内的整体功效。本研究为多基原药材的质量和生物效应等同性共性问题的探索研究提供了借鉴,同时也为车前子及其复方在防治糖尿病的新药开发提供理论依据。

The quality and biological effect of "multi-species" traditional Chinese medicine is a common problem in the process of modernization of traditional Chinese medicine. It is difficult to fully reveal the scientific connotation of the differentiation between varieties only by expounding the differences of chemical components. Plantaginis Semen in Chinese Pharmacopoeia comes from two species. The research team found that there were significant differences in chemical composition and treatment of diabetes between the two species, but the scientific connotation of its research and development is not clear. Based on this, under the guidance of the theory of traditional Chinese medicine, this study takes the two original species of Plantaginis Semen as the research object, and takes the prevention and treatment of metabolic syndrome as the direction, methods and techniques of traditional Chinese medicine chemistry, analytical chemistry, pharmacology of traditional Chinese medicine and metabonomics were used to identify the differences of effective components (groups) by using chemical /pharmacodynamic correlation analysis, and to elaborate the material basis of pharmacodynamic effect and its overall effect in the body of multi base of Plantaginis Semen. This study provides a reference for the exploration and research on the quality and biological effects of multi-source traditional Chinese medicine, it also provides a theoretical basis for the development of new medicines for the prevention and treatment of diabetes by Plantaginis Semen and its compound.
“多基原”中药的品质和生物效应等同性问题是中药现代化进程中普遍关注的难题,而仅仅阐述化学成分的差异性难以全面的揭示品种之间的差异化的科学内涵。基于此,本项目旨在研究多基原车前子车前种子和平车前种子的化学成分、药效及药代动力学特征,探讨其在糖尿病及其并发症治疗中的应用潜力。.通过超高效液相色谱-四极杆飞行时间质谱技术,鉴定了车前子提取物中的61种化学成分,主要包括有机酸类、苯乙醇苷类、黄酮类、胍类衍生物、萜类和脂肪酸类。研究发现,车前种子在苯乙醇苷类和环烯醚萜类成分含量上显著高于平车前种子,而平车前种子在黄酮类成分上更为丰富。.药效学研究表明,车前种子和平车前种子均能显著改善高脂饲料诱导的C57BL/6小鼠的肥胖、高血糖和高血脂症状,且车前种子的整体效果优于平车前种子。此外,车前子对糖尿病肾病小鼠的肾脏病变也有显著改善作用,能够降低血清肌酐、尿蛋白水平,并减少肾脏纤维化。.药代动力学研究表明,车前子中的主要成分如柠檬酸、京尼平苷酸、车前素A等在肝脏、小肠和肾脏中具有较高的浓度,且车前种子组中的药物清除速度较慢,表明其药效持续时间更长。PK/PD模型分析进一步揭示了车前子通过多成分协同作用,调节SOD、TNF-α和胰岛素等药效指标,发挥抗糖尿病和抗炎作用。.本研究明确了车前子的主要活性成分及其在糖尿病治疗中的药效物质基础,为车前子的临床应用和新药开发提供了重要的理论依据和数据支持。研究结果表明,车前子具有显著的降血糖、降血脂和肾脏保护作用,尤其在车前种子中表现更为突出,具有广阔的应用前景。
