Sex determination is an outstanding question in developmental and evolutionary biology. The upstream regulatory mechanisms governing somatic sex determination cascade in insects are highly divergent, however the downstream regulatory mechanisms are relatively conserved. In particular, the tra + tra2 → dsx pathway is fairly common in most insects. In the proceeding phase of this proposed research, we identified tra, tra2 and dsx in the sweet potato whitefly, Bemisia tabaci, and found that dsx and tra2 interacted with each other and were important for male genitalia formation. More importantly, suppression of tra by dietary RNAi down-regulated an array of genes involved in sex determination cascade. Based on our preliminary research, we hypothesized that tra serves as a master regulator in B. tabaci sex determination cascade. To test this overarching hypothesis, we are planning the following studies: 1) RT-PCR and RT-qPCR were used to analyze the splicing and expression patterns of tra and other sex determination genes during early embryonic development; 2) specific RNAi were used to clarify the important role of tra in sex determination of B. tabaci and the relationship between tra and other sex determination genes, so as to further revealing the regulation mechanism of sex determination cascade in B. tabaci. A genetic understanding of tra and its regulatory gene networks in B. tabaci not only will shed light on the sex determination cascade in other haplodiploid insects, but also offer potential molecular targets to control this global invasive insect pest.
昆虫的性别决定存在相对保守的下游调控机制:tra+tra2→dsx→fru。前期研究发现,tra2与dsx之间的相互作用对烟粉虱雄性生殖器的形成很重要,tra在烟粉虱中存在可变剪接,且可能是烟粉虱性别决定级联的主调控因子。本项目在此基础上阐明了tra在烟粉虱性别决定中的关键作用及其调控机理,主要试验结果有:1)利用RT-PCR、RT-qPCR及RNA-seq技术,深入分析了tra及其它性别决定基因在胚胎发育早期的剪接及表达模式,发现早在烟粉虱卵产下的9 h内,tra、dsx、tra2及fru基因就开始表达。tra在受精卵中表达量显著高于未受精卵,而fru在二者间的表达无显著差异。2)tra在烟粉虱中共存在5种剪接体,分别为tra1-tra5。其中,只有tra3能翻译出完整的TRA蛋白质。3)通过GST Pull down试验,证实了BtTRA和BtTRA2蛋白之间的相互作用。4)RNA干扰试验证实,沉默tra,dsx、tra2和fru基因表达量显著下调,说明tra参与调控下游性别决定基因的表达。此外,干扰tra导致雌性烟粉虱腹部末端畸形,卵巢发育受阻。持续干扰影响烟粉虱后代的性比偏移,雌性比例显著下降,说明tra影响了烟粉虱的雌性发育。研究结果不仅为解析其他单双倍体型昆虫的性别决定机制提供了理论依据,还为今后防治烟粉虱提供了潜在的分子靶标。
