树干次生维管组织发育是木材生产的生物学基础,但其精细调控机制目前还很不清楚,尤其是细胞分裂素(Cytokinin, CK)在该过程中的具体作用仍知之甚少。为此,本项目拟以杨树为材料:①克隆次生维管组织特异性启动子以控制报告基因表达,构建各维管组织的转基因报告株系;②采用荧光免疫技术准确表征CK在次生维管组织中的分布模式,搞清CK积累与木材发育之间内在联系;③外施CK和采用转基因技术改变维管形成层中內源CK含量,明确其对于木材发育的调控作用;④利用遗传及生化技术鉴定CK信号通路调控次生维管形成层的下游靶标基因,建立其与CLE-WOX4调控通路之间的联系,阐明CK在次生维管组织中的分子机制;⑤此外,通过组织特异性改变次生维管组织中内源CK和生长素(Auxin)的浓度梯度,探究两者在次生维管组织发育中的协作机制。本项目将为最终揭示林木次生维管发育的分子机理奠定基础,为木材的材性改良提供理论依据。
英文摘要
Wood (also termed secondary xylem) is the most abundant biomass produced by woody plants. Wood development begins with the differentiation of lateral meristem and vascular cambium into secondary xylem mother cells followed by cell expansion, secondary wall deposition, programmed cell death, and finally heartwood formation. In the past decade, significant progress has been achieved in understanding the molecular mechanisms of wood formation in tree species. Hormonal signalling has been shown to play critical roles in vascular cambium cell proliferation. But it is still unclear the regulatory role of cytokinin in the vascular development during wood formation in poplar. In this project, we will isolate these vascular-specific promoters and fuse them into GUS/GFP reporter genes. After transformation, transgenic plants with vascular-specific promoter driven reporter genes will be obtained for exactly distinguishing different vascular tissues. We will further demonstrate the regulation of cytokinin on the differentiation of vascular cambium into xylem mother cells and identify the function of their downstream transcription factors in response to cytokinin signaling in poplar. And also we will figure out the mechanism about cytokinin signalling in phloem transducting into vascular cambium and demonstrate the transcription network encompassing secondary wall regulatory factors and their downstream transcription factors. In addition, we will determine the synergistic regulation of auxin and cytokinin signallings on vascular cambium activity during wood formation. Uncovering the molecular mechanisms of cytokinin regulating vascular development will greatly accelerate our understanding of the molecular control of wood formation in poplar.
次生维管组织是植物重要的物质运输通道,同时也为植物的根茎等组织提供必要的机械支撑,对陆生植物的生长发育起到决定性作用。已有研究表明,在次生维管组织中次生韧皮部是细胞分裂素含量最多的区域。虽然通过改变维管形成层及其周围细胞中细胞分裂素的含量已经证明细胞分裂素对维管组织发育具有至关重要的调控作用,但对于大量聚集在次生韧皮部的细胞分裂素具有何种功能仍待进一步阐明。本项目从细胞分裂素在次生维管组织中的分布模式为切入点,全面解析了大量聚集于次生韧皮部的细胞分裂素调控维管形成层稳态的遗传功能,鉴定了其下游靶基因CLE41/44,阐明了细胞分裂素通过影响TDIF信号从而非细胞自主性调控维管形成层活性的分子机制,并初步揭示了在次生维管组织发育过程中,细胞分裂素对生长素稳态及信号转导的潜在影响。本项目取得的重要结果和关键数据包括:(1)通过免疫荧光技术,精确表征细胞分裂素在次生维管组织中的分布模式;(2)通过建立破坏细胞分裂素分布模式的转基因植株,揭示细胞分裂素分布模式对于维持维管形成层细胞分裂活性是必须的。(3)通过特异性改变次生韧皮部细胞分裂素信号等方式,阐明了定位于次生韧皮部的细胞分裂素信号对维管形成层稳态维持的非细胞自主性调控作用;(4)开展比较转录组分析及生化实验验证,确定了细胞分裂素直接调控CLE41/44基因的转录,并利用遗传回补实验,证实了TDIF信号介导细胞分裂素对维管形成层活性的非细胞自主性调控;(5)利用单细胞测序技术,初步揭示了在次生维管组织发育过程中,细胞分裂素对生长素稳态及信号转导的潜在影响。基于以上结果,我们建立了细胞分裂素信号非细胞自主性维持维管形成层干细胞稳态的分子机制模型,并初步阐述了次生维管组织发育中细胞分裂素对生长素累积及信号转导的潜在调控作用,为解析次生维管组织发育调控机制,完善次生维管组织发育调控网络提供了新线索,为改良木材材料提供了理论依据。
