研究表明NLRP3炎症小体介导固有免疫反应对炎症及相关疾病的发生至关重要,但直接负调节NLRP3分子的机制尚不十分清楚。前期工作发现在多种炎症小体活化刺激下,氨肽酶PSAP可直接结合NLRP3分子,导致其不能寡聚化从而抑制炎症小体复合物的组装活化:提示PSAP可抑制NLRP3炎症小体活化。本项目拟进一步考察氨肽酶PSAP在NLRP3炎症小体活化过程中的动态变化等属性,探讨其在影响NLRP3炎症小体活化中的具体作用和环节;阐明PSAP与NLRP3相互作用的结构域,及二者相互作用对NLRP3分子结构和功能的影响;明确PSAP调节NLRP3分子功能的活性形式;借助可直接靶向PSAP的化合物及PSAP巨噬细胞敲除的小鼠,探讨PSAP介导的调节机制和痛风及肠炎等疾病的相关性及靶向PSAP的治疗学属性。本项目预期阐明NLRP3分子新的调节机制,为相关疾病的临床治疗提供药物靶标。

Studies have shown that the NLRP3 inflammasome mediates the innate immune response is essential for the development of inflammation and related diseases, but the mechanism of direct negative regulation of NLRP3 molecule is not fully understood. Previous work has found that under the stimulation of a variety of inflammasome activation, aminopeptidase PSAP can directly bind to NLRP3 molecule, leading to its inability to oligomerize and inhibit the assembly and activation of inflammasome complex: suggesting that PSAP can inhibit the activation of NLRP3 inflammasome. This project intends to further investigate the dynamic changes of PSAP during the activation process of NLRP3 inflammasome, and explore its specific role and link in influencing the activation of NLRP3 inflammasome; clarify the domain of interaction between PSAP and NLRP3, and how this interaction affect the structure and function of NLRP3; clarifies the active form of PSAP for regulating the function of NLRP3 molecule; By using the compounds that directly target PSAP and PSAP macrophage specific knockout mice and related clinical samples, aiming to explore the relationship between the PSAP-mediated regulatory mechanisms and diseases as well as the therapeutic properties for gout or colitis by targeting PSAP. This project is expected to elucidate the new regulatory mechanisms of NLRP3 molecule and provide drug targets for the clinical treatment of inflammation and related diseases.
NLRP3炎症小体是机体应对危险信号的一个重要分子复合物,它能够引发IL-1β和IL-18的成熟,从而启动天然免疫反应。NLRP3的活化在多种免疫和炎症相关疾病的发展中起着关键作用,因此,研究NLRP3的调控机制对开发新型治疗方法具有重要的临床意义。基于穿心莲内酯能够显著抑制NLRP3炎症小体活化的活性,通过合成生物素偶联的穿心莲内酯,并运用生物素垂钓技术和质谱分析发现穿心莲内酯的靶蛋白是PSAP。进一步通过免疫共沉淀和分子模拟技术,确定了PSAP的结合位点,并发现穿心莲内酯与PSAP的结合不影响PSAP的氨基肽酶活性。通过过表达和敲除PSAP的实验,研究表明PSAP的不同结构域与NF-κB信号通路的活化密切相关。穿心莲内酯能够通过与PSAP的特定结构域结合,阻断PSAP与TRAF6的相互作用,从而抑制NF-κB信号的激活。此外,PSAP还通过不同的泛素化修饰调控TLR4和NLRP3信号通路的传递,进而影响炎症反应。为了进一步验证PSAP在免疫反应中的作用,通过建立巨噬细胞敲除PSAP的小鼠模型,发现PSAP的缺失会导致哮喘、腹膜炎、痛风和银屑病等疾病的加剧,同时NLRP3炎症小体的活化明显增强。这些结果进一步证实了PSAP在调控炎症反应中的核心地位。综上所述,本文揭示了穿心莲内酯通过调节PSAP的泛素化类型,抑制NLRP3信号通路,减轻炎症反应的机制。这一发现为治疗炎症性疾病提供了新的思路,并为开发基于PSAP调控的抗炎药物提供了潜在的目标。此外,穿心莲内酯作为一种天然化合物,凭借其多靶点作用模式,也为中药现代化研究提供了有力的范例,展现了天然药物在多种疾病治疗中的应用前景。
