英文摘要
Microbial symbiosis is the major driving force behind evolution and diversification of eukaryotes. Its role in the evolution of herbivory in animals is currently one of the research hotspots in this field. Due to nitrogen deficiency in plant derived diets, it has been proposed that animals depend on symbiotic microbes to overcome nutrient limitation and explore a plant dietary niche. However, since it is not possible to observe the trophic transition from carnivory to herbivory over a short time scale, there are still no direct evidence to support the contribution of symbiotic microbes in the evolution of animal herbivory. Therefore, it is important to construct a model system to study the role of symbiotic microbes in the early stages of trophic shifts and in the later stages of stable occupation of plant dietary niches. This project aims to test the hypothesis using ants as a study model. We will investigate the contribution of symbiotic microbes in the early stage of trophic shift of invasive ant species. We will further examine whether herbivorous ants harbor stable and host-specific microbiota and address the mechanism behind nitrogen supplementation by symbiotic microbes for herbivorous ant hosts. This study will provide hard evidence on the role of symbiotic microbes in the evolution of ant herbivory and will provide valuable insights on the role of mutualistic interactions in origin and maintenance of ant biodiversity.
共生微生物如何推动真核生物进化及多样性形成是进化生态学领域的前沿研究课题,其中共生微生物对动物的植食性进化的影响是当前该领域的研究热点之一,但是对于这一热点问题尚缺乏系统研究。本项目以蚂蚁为研究模型,通过对10个亚科75个蚂蚁属的969个蚂蚁样品进行16S rRNA定量、扩增子测序、宏基因组分析和操控实验,系统探究宿主植食性饮食习性如何塑造共生微生物的丰度、组成和功能,以及共生微生物如何影响植食性蚂蚁的生态多样性。我们发现不同蚂蚁物种之间的共生细菌丰度和多样性存在很大差异,草食动物中共生细菌的丰度一般较高,而杂食性蚂蚁动物体内共生细菌的数量相比较少。此外,我们发现蚂蚁微生物群中有很大但变化的比例由蚂蚁特化的细菌类群组成。这些细菌通常定殖于多个不同的蚂蚁类群,但在细菌 OTU 水平或更低水平上表现出宿主物种特异性。草食性和肉食性蚂蚁通常拥有更多的细菌,其中特化菌的比例较高,而杂食性蚂蚁的细菌丰度较低。最后,我们通过宏基因组学分析展示了植食性的不同蚂蚁的微生物群在功能上趋同,为宿主提供必要的营养利益。另外,在植食性蚂蚁与微生物建立的共生联盟中,宿主如何维持与微生物间稳定的共生关系是一个巨大的挑战,我们发现,蚂蚁作为社会性昆虫可以通过筑巢蚁后在亲本蚁穴中通过交哺的行为获取共生微生物并传递到新建造的蚁穴中,实现代际间的垂直传递。同时,我们通过以一种植食性龟蚁属蚂蚁作为模式物种,探究了其体内多种共生微生物如何在互惠共生关系中协同合作的问题。通过基因组和代谢组测定,我们发现龟蚁属蚂蚁体内不同共生菌构成的集合体将由中肠共生菌制成的肌苷在后肠中转化为尿素和氨,高效协同地一起为宿主提供食物中缺乏的氮元素。这些结果突出了细菌在蚂蚁生命活动和进化过程中的重要作用,丰富和完善了共生微生物对蚂蚁植食性进化机制的理解。
