顺铂可通过与DNA插层绑定杀死癌细胞,在癌症化疗中有广泛的应用前景。本项目针对顺铂抗癌药物与DNA插层绑定机理这一科学问题,在广义能量片段模型基础上构建溶液环境下DNA片段新模型,采用密度泛函理论(DFT)方法计算DNA低频振动模式,分析DNA低频振动与其内部结构之间的关系,揭示DNA低频振动所具有的生物功能,结合太赫兹波段相干反斯托克斯拉曼散射(THz-CARS)显微镜与低频波段自发拉曼散射技术实验观测DNA溶液的低频振动模式,验证理论模拟结果,以DAPI(二脒基苯基吲哚)与DNA溶液发生的Groove绑定过程为参考实验,最后通过分析对比DNA发生插层绑定与发生Groove绑定前后其低频振动模式变化的区别与内在联系,探究DNA低频振动模式在插层绑定过程中的作用,建立低频振动与DNA插层绑定之间的关系。项目研究结果将为抗癌药物与DNA插层绑定机理研究提供一种新思路和科学依据。

Cisplatin can kill cancer cells by intercalation binding into DNA molecules, and find wide applications in chemotherapeutic therapy of cancers. This research is aimed at elucidating the mechanism of intercalation binding of an anti-cancer drug (cisplatin) into DNA molecule. We will build a new model of DNA fragment in a solution environment based on the generalized energy-based fragmentation. By using density functional theory (DFT) method, we will calculate the low-frequency vibrational modes of the DNA fragments. Based on the calculation we will analyze the relation between the internal structure and the low-frequency vibrations. We will also reveal the corresponding biological functions of the low-frequency vibrations. Combining the terahertz coherent anti-stokes Raman scattering (THz-CARS) microscope and the low-frequency spontaneous Raman scattering technique, we will experimentally observe the low-frequency vibrational modes of DNA solution and verify the theoretical simulations. Using the experiment of Groove binding between DNA and DAPI as a reference, we will explore the role of the low-frequency vibrations of DNA in the intercalation binding between the cisplatin and DNA by analyzing and comparing the distinctions and linkages of the low-frequency vibrational modes changes of the DNA molecules after the intercalation binding and Groove binding. Finally, we will establish a relationship between the low-frequency vibrations of DNA and the mechanism of the intercalation binding. The research results will provide a new idea and a scientific basis for the research on the mechanism of intercalation binding between the anti-cancer drug and DNA molecule.
针对顺铂与DNA插层绑定这一科学问题,本项目在理论上考虑了溶液环境中的氢键作用,以此构建了DNA溶液模型,并利用密度泛函理论计算了DNA低频振动模式;在实验上利用相干反斯托克斯拉曼散射(CARS)显微系统测量DNA低频振动模式,结合数值模拟与实验测量阐明了DNA与顺铂插层绑定机理。在ACS Photonics, 11, 3467−3475(2024)的成果中,实验证明了电子共振增强作用下的CARS显微术,并基于这种CARS显微术揭示了细胞和生物组织中被染色蛋白的空间分布规律;在Molecules, 26, 5178(2021)的成果中,提出了一种近简并四波混频显微术的简化方案(SNDFWM),并证明了其在生物组织和纳米材料方面的高灵敏、高空间分辨率的能力;在物理学报, 73, 164207(2024)的成果中,从实验和模拟仿真上基于二次谐波产生光谱与显微成像阐明了纳米线空间精密定位的原理;在Physical Review Applied, 16, 044013 (2021)的成果中,我们报道了一种无需传统强度关联计算的光散射成像方法,并揭示了这种成像方法在目标物体识别方面的成像机理。总结以上的四篇重要成果:创新的将电子共振效应耦合到CARS光谱与显微成像研究中,实现了对细胞及生物组织中蛋白质的超灵敏探测,为高度多重选择性成像提供了途径。其次,提出的SNDFWM显微镜能够在体内纳米药物追踪方面找到潜在重要应用。此外,SHG光谱与显微成像在纳米材料空间高精度定位研究为纳米材料在生物医学方面的潜在应用提供了重要启示。最后,热光“鬼”成像研究为在低光或光子计数条件下以及更宽光谱范围内使用二元探测器进行鬼成像提供了一种潜在途径。申请人在项目执行过程中,总共发表5篇SCI论文,申请3项发明专利(1件授权),授权1件软件著作权,相关研究成果分别荣获陕西高等学校优秀科学技术成果二等奖和陕西省自然科学优秀学术论文三等奖。
