以羊痘病毒为代表的DNA病毒感染在陕北地区羊常发病中占极高比例,已成为制约该地区养羊业发展的瓶颈。本研究在利用Lenti-XTMTet-Off系统构建cGAS及PGAM5过表达细胞系,和利用CRISPR/Cas9技术构建cGAS及PGAM5基因敲除细胞系基础上,采用免疫荧光、免疫共沉淀和激光共聚焦等方法研究PGAM5与cGAS互作并确定作用最佳条件,然后利用Western blot、qPCR、TCID50测定和ELISA等技术研究cGAS-STING通路在宿主细胞抗CPV感染的作用,及CPV感染后PGAM5对cGAS-STING通路的调控作用,最终明确PGAM5调控cGAS-STING通路抗CPV感染的分子机制。本项目的开展可为新型广谱抗DNA病毒疫苗及药物的设计提供重要的科学依据,同时也将为陕北地区羊痘等DNA病毒病的防控提供理论支撑,从而为该地区养羊业快速发展保驾护航。

The infection of DNA virus, represented by Capripoxvirus, accounts for a very high proportion of sheep epidemics in northern shanxi, which has become a bottleneck problem restricting the development of sheep industry in this region. In the present study, based on the construction of cGAS and PGAM5 overexpressed cell lines by Lenti-XTMTet-Off induced expression system and the construction of cGAS and PGAM5 knockout cell lines by CRISPR/Cas9 system, the protein interaction between PGAM5 and cGAS was studied by immunofluorescence, immunoprecipitation and laser confocal methods and the optimal conditions were determined. And using Western blot, Real-time PCR, determination of TCID50 and ELISA technology, the role of cGAS-STING signaling pathways in the host cell against CPV virus infection and the regulation of cGAS-STING pathway by PGAM5 after CPV infection were studied, which help to ultimately clarify the molecular mechanism of PGAM5 regulation of cGAS-STING pathway against CPV infection. The smooth development of the project will provide an important scientific basis for the design of new broad-spectrum anti-DNA virus vaccine and anti-DNA virus drugs, and will also contribute to the prevention and control of DNA virus diseases such as sheep pox and the rapid development of sheep industry in Northern Shanxi.
以痘病毒为代表的DNA病毒对规模化养殖业危害极大,为了探寻机体抗病毒的分子机制,本研究通过qRT-PCR和Western-blot技术研究PGAMs在不同组织和细胞中的表达谱,结果表明PGAMs家族成员主要在心脏、肾脏、淋巴结组织中表达水平高;通过激光共聚焦显微镜对其细胞和亚细胞定位进行研究,结果表明PGAMs主要定位于细胞质的高尔基体、内质网,痘病毒感染后其亚细胞定位没有发生变化;通过GST-pull down和Co-IP技术研究PGAMs与cGAS、STING的相互作用,结果表明PGAM1、PGAM2、PGAM4与cGAS蛋白能够相互作用,而PGAM1、PGAM2和PGAM4与STING蛋白没有相互作用,PGAM5与cGAS和STING均能相互作用;以Hela细胞为研究对象,通过激光共聚焦显微镜证明了外源性PGAM5在线粒体的亚细胞定位;以MDBK细胞为研究对象验证了内源性PGAM5,感染和非感染状态下都定位于线粒体,且痘病毒感染后PGAM5线粒体的定位不发生改变;信号通路研究结果表明,痘病毒感染后能通过PGAM5激活cGAS-STING信号通路,且随着痘病毒感染时间和感染强度的增加,对下游关键分子及干扰素的诱导能力增强;基因工程技术表达纯化的gcGAS酶蛋白,能够体外酶促催化合成山羊源2′ 3′- cGAMP,并能诱导靶细胞分泌IFN-β和TNF-α,并在体外发挥抑制病毒增殖的作用,具有良好的抗病毒效果。本项目的开展为新型广谱抗DNA病毒疫苗及药物的设计提供重要的科学依据,同时也为DNA病毒病的防控提供理论支撑,从而为我国畜牧业的健康快速发展保驾护航。
