皮层中间神经元在神经环路中闸控着信息的传递和整合,其形态、电生理特征、分子标记物及功能各异,具有高度异质性,包含基本不交叉的三大亚群,即PV+、SST+和VIP+神经元。不同亚群的发育缺陷常导致多种脑高级功能障碍,疾病的临床症状亦不尽相同,但机制不明。我们前期利用Pv-Cre、Sst-Cre、Vip-Cre分别在不同亚群中敲除转录因子Foxg1,发现PV+神经元形成的突触数目减少,SST+神经元亚型分化异常,伴随着小鼠焦虑和社会交往能力的改变。本项目将在此基础上通过形态学和电生理手段明确Foxg1对三大亚群神经元分化成熟的调控以及对皮层神经环路兴奋性和抑制性平衡的影响;利用行为学手段比较分析不同亚群缺陷对小鼠行为的影响,进而解析FOXG1综合征认知和情感障碍、癫痫等症状的细胞机制;采用CHIP-Seq和RNA-Seq探讨其机制。本项目旨在理解各亚群的分化调控机制和各亚群在脑高级功能中的作用

Although only consisted of 20-30% in the cortex, GABAergic inhibitory interneurons play critical roles to gate signal flow and sculpt network dynamics. Cortical inhibitory interneurons are a highly heterogeneous population and consist of several subpopulations that exhibit distinct cellular properties, including specific morphologies, molecular profiles and electrophysiological properties. Developmental defects of interneurons often lead to cognitive and emotional defects as well as limitation of social ability. It has been demonstrated that the calcium-binding protein parvalbumin (PV), the neuropeptide somatostatin (SST), and the vesointestinal peptide (VIP) are expressed in largely non-overlapping subpopulations in the cortex. During development, most cortical interneurons are derived from the ganglionic eminence and tangentially migrate to the dorsal cortex. Subpopulations of PV+ and SST+ interneurons originate from the medial ganglionic eminence, while the VIP+ interneurons derives from the caudal ganglionic eminence. However, the mechanisms underlying subpopulation differentiation of interneurons remain unclear. In this study by conditional disruption of Foxg1 in distinct subpopulations achieved by crossing Pv-Cre, Sst-Cre, Vip-Cre with Foxg1fl/fl, respectively, we have found the number of inhibitory synapses is significantly reduced in Pv-Cre;Foxg1fl/fl mice, and the subtype specification of SST+ interneurons is affected in Sst-Cre;Foxg1fl/fl. Moreover the Pv-Cre;Foxg1fl/fl mice exhibit increased anxiety and abnormal social behaviours. Next, we will assess the effects of Foxg1 deletion on each subpopulation differentiation including morphological changes and their electrophysiological properties. Behavioural tests will be performed on Pv-Cre; Foxg1fl/fl, Sst-Cre; Foxg1fl/fl, and Vip-Cre;Foxg1fl/fl mice, respectively, to further compare the contributions of each distinct subpopulation to the cognitive, emotional and social abnormalities. CHIP-Seq and RNA-seq will be employed to explore downstream targets of Foxg1 to regulate the differentiation of subpopulation interneurons. Our study will put insights into the mechanisms of the development of cortical interneurons and FOXG1-related disorders.
皮层中间神经元在神经环路中闸控着信息的传递和整合,其形态、电生理特征、分子标记物及功能各异,具有高度异质性,包含基本不交叉的三大类群,即PV+、SST+和5-HT3aR+/VIP+神经元(VIP+神经元亚群包含在5-HT3aR+类群之内)。不同类群的发育缺陷常导致多种脑高级功能障碍,但机制不明。转录因子FOXG1是调控端脑发育的一个重要因子,FOXG1基因突变的患者表现出焦虑抑郁、精神分类症、高度活跃、高度焦虑等不同症状,但是发生机制有待于解析。本项目利用细胞特异性SST-Cre、PV-Cre、5HT3aR–Cre工具小鼠和Foxg1fl/fl小鼠,分别在SST+、PV+和5HT3aR+不同类群中间神经元中敲除FOXG1,旨在明确FOXG1对中间神经元分化、成熟的调控机制,探讨不同类群中间神经元发育异常与FOXG1综合征发病机理。..我们发现SST+中间神经元缺失Foxg1导致SST+中间神经元数量明显减少且定位异常,亚型如SST+Reelin+、SST+Calretin+、SST+NPY+等命运发生了改变,SST-Cre;Foxg1 cKO小鼠表现出认知缺陷、社会交往障碍以及抑郁样行为等,低剂量氯硝安定处理可挽救小鼠抑郁样行为。PV+神经元缺失FOXG1导致树突复杂性降低,突触减少。PV-Cre;Foxg1 cKO小鼠表现为感觉门控障碍,前脉冲抑制受损等精神分裂症样行为,可部分模拟FOXG1突变引发精神分裂症状的表型。进一步研究发现FOXG1直接调控II类代谢性谷氨酸受体Grm3的转录,进而影响PV+神经元突触传递,影响神经环路的E/I平衡。5-HT3aR+神经元缺失FOXG1后,5-HT3aR+抑制性中间神经元的树突复杂性增加, 命运特化异常。5-HT3aR;Foxg1 cKO小鼠处于高度的焦虑状态和高度活跃状态,我们进一步发现FOXG1可通过直接调控reelin和pten影响5-HT3aR+神经元的发育和功能。.本项目研究表明FOXG1在SST+、PV+和5HT3aR+的三个类群GABA能中间神经元中作用和调控机制各不相同,各亚群发育异常所致脑高级功能障碍也不尽相同。我们的研究揭示了不同类群GABA能中间神经元异常导致FOXG1综合征患者焦虑抑郁以及高度活跃等不同的临床症状的发生机制,为阐明FOXG1综合症以及相关疾病的发生机制、探讨新的干预手段提供了新的思路
