微藻是类胡萝卜素、油脂等高附加值产物的重要来源,应用前景广阔。微藻在积累高值组分同时常伴随大量廉价有机物如碳水化合物、蛋白质的合成,这四类有机物是微藻胞内碳存在的主要形式,各自合成存在相互影响、共同竞争碳源底物现象。由于对微藻胞内碳分配代谢机制不清,导致微藻高值组分生产不够稳定、产率不高,成为影响微藻产业化发展的主要难点之一。本项目突破现有研究均以藻群为对象,存在细胞异质性被忽略、分析步骤复杂繁琐造成的样品损失和结论失真等问题,创新性地将显微拉曼光谱技术与单细胞生化分析技术有机结合,利用显微拉曼光谱仪实现单细胞内四类有机物成像分析,利用单细胞仪实现痕量碳代谢中间产物单分子特异性快速检测,两者优势互补,可有效追踪不同LED光质下微藻胞内碳分流、储运及互作关系,揭示微藻单细胞碳分配机制,并最终获得微藻单细胞高效优质生产的优化光质方案。以期通过提高微藻个体细胞产能,从根本上实现藻群的稳定高产。
英文摘要
Microalgae, an important source of high-value organic compounds such as carotenoids and lipids, have broad application prospects. Large amounts of low-value organic compounds (e.g. carbohydrates, proteins) are formed during the accumulation of high-value products in microalgae. These four kinds of substances are the main forms of intracellular carbon in microalgae, and their anabolisms affect each other and compete for carbon substrate. Little is known about the mechanisms determining the synthesis and metabolism of organic products and carbon partitioning in microalgae, which leads to the unstable and low yield of high-value products of microalgae. That is one of the main difficulties that influence the development of microalgae industrialization. This project breaks through the limitations of the present studies, which all focus on microalgal population, have problems such as ignorance of cell heterogeneity and the loss of samples due to the complexity of analysis steps, and thereby cause the distortion of conclusions. By using the micro-Raman spectroscopy which is suitable for the imaging analysis of organic products in single microalgal cells, combining with the single cell analyzer which is an ideal means in detection of trace intermediates of carbon metabolism, to study the distribution, storage, transportation and interaction of microalgal intracellular carbon under different LED light quality, to reveal the mechanisms of intracellular carbon partition of microalgae, thereby to develop an optimized artificial supplementary lighting for high efficiency and quality production of single microalgal cells. The project is expected to achieve stable and high yield of microalgal population by improving individual cell productivity.
微藻广泛应用于食品、医药、饲料等领域,也是各国积极发展的可再生能源原料。微藻在积累高附加值组分(如油脂、类胡萝卜素)的同时,常伴随大量廉价有机物(如碳水化合物)的合成,这些物质均是微藻胞内碳存在的主要形式,各自合成存在相互影响、共同竞争碳源底物现象。由于对微藻生长过程中胞内有机产物的合成代谢规律及碳分配机制不清,导致许多微藻高值组分转化率较低、生产不够稳定,严重影响了微藻的产业化发展。本项目利用单细胞纳米探针结合多尺度光谱成像技术,系统开展了不同LED光质调控微藻单细胞碳分配机制研究工作。研究发现,相较于大多数研究中使用的单侧光布置,环绕式光源更利于微藻对光能的利用,且光能损耗小、稳定性好。研发了基于适配体/有机基团修饰的纳米探针,可不经处理直接内化入微藻活体细胞,实现对微藻细胞内关键碳代谢中间产物(ATP、NADH、丙酮酸等)的高灵敏、特异性检测;构建了微藻细胞和群体尺度的有机产物快速检测方法。基于以上方法,揭示了微藻(棕鞭藻)单细胞碳代谢中间产物与下游有机产物(碳水化合物、油脂和蛋白质)互作关系和机制。结果表明,LED蓝光通过提高棕鞭藻细胞中ATP和NADH水平,优先驱动碳水化合物、蛋白质和光保护色素(类胡萝卜素)的合成,保障微藻的快速增殖。丙酮酸作为油脂和碳水化合物代谢的交汇点,红光通过提高丙酮酸转化,增加油脂合成的碳骨架流,促进油脂合成。需要说明的是,棕鞭藻(金藻)藻液几乎透明的特点使得光源在其中的穿透性强,与陆生植物接受光照的特性接近,故棕鞭藻的碳代谢研究结论对陆生植物的光源调控具有很好指导;但对于光透过性很弱的绿藻(如蛋白核小球藻)藻液,光源在其中易发生散射、反射现象使波长红移,导致研究结果与棕鞭藻差异较大:LED绿光或蓝光能显著促进蛋白核小球藻生物量增加,蓝光可显著提升油脂累积,而红光培养可促进微藻中碳水化合物合成。因此,在微藻的工厂化养殖中,需针对不同微藻特征和目标产物生产需求,选择相应的LED光质来定向诱导微藻细胞中碳代谢中间产物的转化,从而促进下游目标有机产物的合成。本项目不仅为光源调控微藻等植物的细胞-群体多尺度碳分配机制研究提供了新的思路和方法,也为植物高效优质生产的光源调控方案设计提供了理论支撑,具有重要的实践价值和应用潜力。
