英文摘要
Melatonin is a key hormone in maintaining the circadian rhythms of organisms. In vertebrates, it is mainly synthesized and secreted by the pineal gland. Previous studies of the applicant have proved that microRNA-7 plays an important role in regulating the synthesis of melatonin in pig pineal gland. However, the related molecular mechanisms have not been clarified clearly. Basing on the previous researches by the applicant, this project is thus proposed to: Investigate the relationship between the rhythmic synthesis of melatonin and the spatiotemporal expression of microRNA-7; Identify and research the upstream signal transduction pathway as well as downstream target genes and/or molecular intersections of microRNA-7 in regulating melatonin circadian rhythm; Reveal the molecular network mechanism of microRNA-7 regulating melatonin synthesis. These results are critical for understanding the regulatory mechanisms of circadian melatonin synthesis, and will have important guiding significance for the healthy breeding of livestock including pigs.
褪黑素是维持生物体昼夜节律的关键激素。在课题组前期研究中,已经证明microRNA-7(miR-7)通过抑制褪黑素合成关键酶Aanat表达从而抑制猪松果体褪黑素合成,但相关的分子机制仍未阐明。在本项目中,利用生物信息学手段对miR-7调控褪黑素合成的下游靶基因进行筛选,并检测筛选获得的基因在猪松果体内的表达模式,发现生物钟基因Period2(Per2)以及m6A甲基转移酶/去甲基化酶在猪松果体内的表达呈现明显的昼夜节律性模式,与miR-7以及Aanat的表达变化有显著的相关性。进一步研究其在松果体褪黑素合成中的功能,发现抑制m6A甲基化导致猪松果体褪黑素合成增加。此外,在项目进展过程中,发现miR-7能够调节垂体发育,并鉴定Ckap4和Gli3为miR-7下游直接靶基因。综上,本项目通过对miR-7靶基因的筛选,拓展了松果体褪黑素合成以及垂体发育调控的分子网络,为该领域进一步的研究提供了重要的理论基础。
