内生菌的跨植物世代传播能够维系植物-微生物共生体的连续性,这种传播方式对植物的生态、健康和生产力有深远影响。现阶段关于植物内生菌跨世代传播的研究都集中在种子的垂直传播途径上,尚不清楚无性繁殖植物中内生菌的垂直传播规律,并且缺乏对植物内生菌群落组成及传播机制的系统研究。克隆植物活血丹(Glechoma longituba)以匍匐茎连接无性繁殖的子代分株,所形成的分株相连网络体系能够观察到内生菌跨多级世代的连续传播过程,可作为研究植物内生菌跨世代传播的一个理想材料。本项目拟以活血丹叶、根和匍匐茎中的内生菌为研究对象,运用高通量扩增子测序、传统培养和96孔板液体高通量培养技术,研究内生菌的群落组成和动态变化,结合生物信息学分析手段,探究影响内生菌群传播的因素和发生的传播过程,进而阐明植物内生菌的传播策略和机制,为深入理解植物-微生物多方互作机制和内生菌共生体的进化适应提供新见解。

The transmission of endophytes across plant generations can maintain the continuity of plant-microbial symbiont, which has a profound impact on the ecology, health, and productivity of plants. At present, most of the studies on endophytes transmission across plant generations have focused on the vertical transmission of seeds. However, it is still unclear about the vertical transmission pattern of plant endophytes in clonal plant. In addition, there is still a lack of systematic research on the composition of plant endophytes and transmission mechanisms. The clonal network system formed by the clonal plant Glechoma longituba can observe the continuity of the microbial community transmission to the plant progeny. It is a suitable material to study the transmission mode and mechanisms of endophytes across multi-generations. This project will investigate the endophytic microbes in the phyllosphere, roots, and stolon of the clonal plant Glechoma longituba. Metagenomics study with high-throughput sequencing techniques, culture-dependent, and high-throughput cultivation methods will apply to study the composition and dynamic changes of endophytic microbial communities that transmit across plant generations. Combined with the bioinformatics analysis, analyzing the factors that affect the transmission of endophytes and the assembly processes that occur. Consequently, the transmission strategies and mechanisms of plant endophytes will be illustrated. Knowledge of this will provide a new insight for understanding the mechanism of plant-microorganism interactions and the evolutionary adaptation of endophytic symbiont.
揭示内生菌介导的生物过程是研究植物适应与进化的重要途径,对增强生态系统稳定性和促进生物多样性具有重要意义。内生菌在世代间的垂直传递对于植物后代的发育起着关键作用。与种子植物不同,无性繁殖植物缺乏种子这一垂直传递的媒介,目前的研究表明它们仅依赖于营养结构进行内生菌的垂直传递。然而,目前尚不清楚内生菌在无性繁殖植物中的垂直传递模式、动态变化,以及在植物繁殖过程中这些变化对后代的影响。为了解决上述问题,我们以活血丹(Glechoma longituba)为研究对象,对内生菌介导的在克隆植物中的生物过程进行了深入研究。研究揭示了内生菌在克隆植物中垂直传递的新机制:(1) 匍匐茎作为内生菌传递的通道,表现出随距离递减的特性;(2) 内生菌群落结构在代际间趋于收敛,变得更加简单但更稳定,由广适性物种占主导地位;(3) 垂直传递的内生菌保留了对后代生长和健康至关重要的功能特性。这些发现支持了这样一种假设:无性繁殖植物的垂直传递受到母体供应和后代选择的双重调控,通过缓冲匍匐茎中的距离递减效应,为后代建立所需的内生菌群落。总体而言,该研究阐明了无性繁殖植物中内生菌垂直传递的机制,并进一步丰富和深化了对宿主进化潜力和宿主-微生物群共进化的理解。
