肾移植是治疗终末期肾病的首选方法,但抗体介导的移植肾排斥反应仍是导致移植肾功能丧失的重要因素。我们前期利用小鼠肾移植和动脉移植分别构建的急、慢性排斥模型,均证实triptolide可显著抑制抗体的生成,但机制不明。现有研究发现B细胞身份基因相关的超级增强子(Super-enhancer),在抗体生成中扮演关键角色,而最新研究证实triptolid可调控Super-enhancer,另外,预实验中我们发现triptolide可调控B细胞身份基因的表达。据此,我们提出triptolide通过调控B细胞身份基因相关的Super-enhancer抑制抗体生成的科学假说。本项目拟在分子、细胞和动物组织三个层面,利用ChIP-seq技术、小鼠肾移植模型等解析Triptolide调控Super-enhancer抑制B细胞抗体产生的关键分子机制。项目研究成果有望为抗体介导移植肾排斥的防治提供新途径。
英文摘要
Kidney transplantation is the most ideal way to treat patients with end-stage renal disease. However, antibody-mediated rejection of the transplanted kidney is still an important factor leading to the loss of graft function. We previously used mouse kidney transplantation and arterial transplantation to construct acute and chronic rejection models and found that triptolide could significantly inhibit the production of antibodies, but the mechanism is unclear. Studies have confirmed that super-enhancers related to B cell identity genes play a key role in antibody production, and triptolid can regulate the expression of super-enhancers. In addition, in preliminary experiments, we found that triptolide can regulate the expression of B cell identity genes. Therefore, we put forward the scientific hypothesis that triptolide inhibits antibody production by regulating the super-enhancer related to B cell identity genes. This project intends to use ChIP-seq technology and mouse kidney transplantation models to analyze the key molecular mechanism of Triptolide's regulation of super-enhancer to inhibit the production of antibodies at the three levels: molecules, cells and animal tissues. The research results of the project are expected to provide a new method for the prevention and treatment of antibody-mediated renal transplant rejection.
肾移植是解决终末期肾病的最佳途径。但移植术后抗体介导的排斥反应(Antibody-mediated rejection,AMR)的治疗难度大,是导致移植肾远期功能丧失的主要原因之一。现有治疗手段未能很好的改善AMR的预后,临床上迫切需要一种新的方法防治AMR,改善移植肾的预后。本项目发现了调控B淋巴细胞身份的关键基因的Super-enhancer,在分子水平,证实了Triptolide通过调控B淋巴细胞身份的关键基因的Super-enhancer抑制抗体生成的分子机制,项目的顺利进行,有望开发一种新的药物能调控B细胞产生抗体的关键环节,并从源头上抑制抗体的生成,从而减少供体特异性抗体(donor specific antibody, DSA)的形成,预防AMR的发生,改善移植肾的远期预后,具有重要的临床意义。其次,我们发现TSPO配体通过影响B细胞代谢障碍改善移植术后抗体介导的排斥反应;机制上我们发现TSPO配体体外抑制B细胞糖酵解和线粒体呼吸水平,同时抑制B细胞代谢相关分子表达水平。该研究的顺利发表为临床AMR的防治提供新的治疗策略。最后,我们发现了一种靶向器官的免疫诱导新方案;我们自主研发了一种同时具备锚定细胞表面和保留PD-L1生物学功能的新型重组蛋白map-PD-L1。利用大鼠肾移植模型,证实离体阶段map-PD-L1可锚定修饰供肾,使得供肾血管内皮细胞表面高表达PD-L1,进而减少移植肾局部T细胞的浸润,同时可促进Treg细胞在供肾局部的分化,从而改善移植术后排斥反应介导的移植肾损伤;该研究充分利用供体器官离体阶段的独特特点,对供体器官进行特异性锚定修饰,为器官安装好“防弹衣”,从而有效避免全身应用带来的风险,有望为器官移植术后提供一种靶向器官的免疫诱导新方案,实现移植受者的精准治疗,具有重要的临床转化前景及社会意义。
