肠黏膜上皮屏障在维持机体健康中起着关键性作用,其功能损伤与肥胖、糖尿病等多种疾病发生和发展密切相关。研究显示,膳食多酚类化合物能改善肠黏膜上皮屏障功能,但其作用机制不清楚。课题组前期研究显示猕猴桃多酚能缓解高脂膳食造成的肠道通透性增加,提高肠黏膜紧密连接蛋白mRNA的表达和线粒体呼吸链复合酶的活性,其对肠黏膜上皮屏障功能损伤是否与线粒体功能的恢复有关并不清楚。鉴于线粒体自噬在维持线粒体功能中的关键作用,本项目假设猕猴桃多酚能通过促进线粒体自噬改善肠黏膜屏障功能。项目拟通过动物实验结合细胞培养,运用形态学、免疫荧光组织化学、流式细胞术、蛋白质印记等技术,研究猕猴桃多酚对肠黏膜上皮紧密连接蛋白分布、线粒体功能的影响,并从AMPK-ULK自噬途径探索猕猴桃多酚对肠黏膜上皮屏障功能保护作用的机理。该项目的完成为膳食多酚预防与肠黏膜损伤有关疾病的发生,促进机体健康,提供理论支撑。
英文摘要
Intestinal epithelial barrier plays a key role in maintaining body health, and its impairment is related to the occurrence and development of many diseases, including obesity and diabetes. Recent studies have shown that dietary polyphenols may improve the function of intestinal barrier, but the mechanism is still unclear. The previous research results of the our group showed that kiwifruit polyphenols can alleviate the increase of intestinal permeability caused by high-fat diet, and improve the expression of mRNA in tight junction protein and the enzymatic activity of mitochondrial respiratory chain complex, but we don’t know whether the mitigative effect of kiwifruit polyphenols on intestinal barrier function is related to the recovery of mitochondrial function. Given the critical role of mitophagy in maintaining mitochondrial function, this project hypothesized that kiwifruit polyphenols could improve intestinal barrier function by promoting mitophagy. The project intends to study the effects of kiwifruit polyphenols on the distribution of tight junction proteins and mitochondrial function in intestinal epithelium cell, and explore the protective effect and mechanism of kiwifruit polyphenols on intestinal barrier function based on AMPK-ULK signal pathway of mitophagy,through animal experiments combined with cell culture, using morphology, immunofluorescence histochemistry, flow cytometry, protein imprinting and other techniques. So, the completion of this project can provide theoretical support for the prevention of diseases related to intestinal mucosal injury by dietary polyphenols and the promotion of health.
肠黏膜上皮屏障在维持机体健康中起着关键性作用,其功能损伤与肥胖、糖尿病等多种疾病发生和发展密切相关。同时,肠黏膜上皮细胞是机体过更新最快的组织细胞,能量需求极高。研究显示肠道的能量需求占机体总能耗的20%-30%,充足的能量供应是维持肠上皮细胞紧密连接的物质基础。猕猴桃在传统药物典籍中记载能对消化系统疾病有一定的缓解作用。因此,本项目立足与线粒体结构和功能的稳态,研究猕猴桃多酚对肠黏膜上皮细胞屏障功能的保护作用。.本项目从细胞和动物水平对其作用机制进行探讨,采用脂多糖诱导肠道屏障功能障碍。研究结果显示,猕猴桃多酚能抑制脂多糖造成的Caco-2通透性增加,表现为FD-4透过率下降和TER上升,以及紧密连接蛋白mRNA表达的升高。通过蛋白质组学对其作用机制进行初步筛选,发现猕猴桃多酚干预后上调53个蛋白和下调55个蛋白,主要影响了氧化磷酸化、线粒体自噬、三羧酸循环、紧密连接及AMPK信号途径等。这一结果预示着线粒体可能是猕猴桃多酚缓解脂多糖造成Caco-2细胞损伤的关键靶点。通过对活性氧自由基、膜电位、ATP含量、线粒体呼吸链复合酶的活性,显示猕猴桃多酚干预线粒体的损伤得到缓解,ATP含量升高。针对线粒体稳态调节的关键蛋白mRNA的表达,发现AMPK、ULK-2变化较为明显,同时自噬荧光数据显示,猕猴桃多酚干预线粒体自噬程度减轻。这暗示着AMPK/ULK2介导的线粒体自噬途径可能是猕猴桃多酚缓解脂多糖造成损伤的一个重要途径。采用高浓度的AMPK激动剂AICAR刺激细胞,猕猴桃多酚干预能降低AMPK/ULK2的表达,并抑制线粒体自噬和肠道通透性下降。最后动物实验结果表明,猕猴桃多酚干预能提高结肠上皮细胞紧密连接蛋白mRNA表达,提高结肠的能荷水平升高,同时线粒体结构趋于正常,线粒体自噬小体减少,AMPK、ULK2 mRNA水平和蛋白水平的表达均下降。综上实验表面猕猴桃多酚能通过抑制AMPK/ULK2线粒体自噬缓解脂多糖造成的肠道通透性增加。.该研究培养了硕士研究生4名,发表文章6篇,其中一区论文3篇,培养青年教师2名,参与学术会议6人次。
