Cigarette smoke (CS) exposure is the most important risk factor for chronic obstructive pulmonary disease (COPD). The applicant previously discovered that neutrophil extracellular traps (NETs) are important mediators of CS causing COPD, but how it regulates the formation of NETs is still unclear. Given the "bridge" role of m6A modification in the pathogenesis of environmental factors, the applicant found through bioinformatics analysis and pre-experiments that the methylase ALKBH5 was down-regulated in neutrophils stimulated by sputum and CS in COPD patients while reading protein IGF2BP2 and autophagy-related gene PLAC8 were up-regulated. Besides, ALKBH5 could reduce the m6A methylation modification and expression level of PLAC8. Thus, it is proposed that CS-mediated ALKBH5 induces PLAC8 m6A modification, regulates the expression of the reading protein IGF2BP2, enhances the mRNA stability of PLAC8, jointly promoting the formation of NETs mediated by autophagy, promoting the airway inflammation, and accelerating COPD progression. This project intends to focus on ALKBH5-mediated PLAC8 m6A methylation, revealing the mechanism of CS promoting the formation of NETs and the process of COPD from multiple dimensions such as clinical, animal, cell, and molecule, and provide new ideas for developing COPD treatment strategies targeting NETs in the future.
本研究旨在深入探讨香烟烟雾(CS)诱导的慢性阻塞性肺疾病(COPD)的发病机制,并特别关注m6A RNA甲基化在这一过程中的作用。中性粒细胞胞外诱捕网(NETs)作为CS致病的关键介质,其调控机制的研究对于理解COPD的发病过程具有重要意义。本研究通过生物信息学分析和实验验证,发现COPD患者诱导痰和CS刺激的中性粒细胞中,甲基化酶ALKBH5的表达显著下调,而阅读蛋白IGF2BP2和自噬相关基因PLAC8的表达则上调。这一发现提示我们,ALKBH5可能在调控PLAC8 m6A甲基化和表达水平中起到重要作用,进而影响NETs的形成和COPD的进程。并通过实验观察到CS刺激后,中性粒细胞中ALKBH5的表达下调,而PLAC8的m6A甲基化水平则上调。同时,我们还发现ALKBH5能够降低PLAC8的m6A甲基化和表达水平,进而调控细胞自噬和NETs的形成。同时构建了COPD小鼠模型,并观察到ALKBH5表达下调、PLAC8表达上调以及NETs形成增加的现象,进一步验证了我们的假设。此外,本研究还关注了其他m6A甲基化相关酶在COPD中的作用。我们发现ZC3H13作为一种m6A甲基化转移酶,在COPD中差异表达,并通过m6A修饰增强ITGA6的表达和mRNA稳定性,影响支气管上皮细胞炎症和纤维化,进一步揭示了m6A甲基化在COPD发病机制中的重要性。综上所述,本研究从临床、动物、细胞、分子多维度揭示了CS促进NETs形成和COPD进程的机制,为今后靶向NETs的COPD治疗策略开发提供了新思路。同时,本研究还阐明了m6A去甲基化酶在COPD中的重要作用,为理解COPD发病机制和从m6A甲基化角度开发靶向治疗提供了新的途径,有望为COPD的治疗带来新的突破。
