群体中不同性别个体的存在不仅对其生理发育有调节作用,而且对衰老和寿命有着重要影响。但是这种两性对抗行为对个体代谢与衰老的调控机制目前还知道甚少。我们发现模式动物秀丽隐杆线虫的雄性个体特异分泌蛋白F56D2.8能够通过精液进入到雌雄同体个体内,并作用于肠道细胞驱动其衰老机制减短其寿命。以上预实验结果提示分泌蛋白F56D2.8参与雄虫特有的促进雌雄同体衰老的功能机制。本项目拟利用蛋白纯化、RNAi反向遗传学筛选和转录组对比分析来探讨分泌蛋白F56D2.8对线虫肠道细胞的调节衰老的作用受体和信号通路,并且通过的分析蛋白质的保守性功能区间利用小鼠模型来研究其对哺乳动物的凋亡与代谢功能机制。本项目将为交配行为对动物个体的衰老和寿命的影响作用机制提供新的理论假说。
英文摘要
The existence of different genders in the group of organism not only has a regulatory effect on their physiological development, but also has an important impact on aging and longevity. However, the mechanism of sexual conflict that acts on individual metabolism and aging is still poorly understood. We found that the specific secretory protein F56D2.8 of male individuals of the model animal Caenorhabditis elegans can enter into hermaphrodite individual through seminal fluid and act on intestinal cell to drive its aging mechanism to shorten its lifespan. The above pre-experimental results suggest that the secreted protein F56D2.8 participates in the unique functional mechanism of male and hermaphrodite aging regulation. This project intends to use protein purification, RNAi reverse genetics screening and transcriptome comparative analysis to explore the role of secreted protein F56D2.8 on intestinal cells to regulate aging. At the same time, this project intends to use cell line and mice model to study its mechanism of apoptosis on mammalian cell and the metabolic function on mice. This project will help to provide a new theoretical hypothesis for the effect of mating on the individual's aging and longevity.
随着中国人口结构的改变,社会步入老年化阶段,研究衰老的调节因素,改善国民老年人生活质量是国家健康重大战略之一。模式动物的研究揭示了许多衰老和寿命调控的机制与细节,但是异性个体的存在和相互行为对衰老和寿命调控的关键因子还没有被发现。本项目通过模式动物线虫的寿命实验方法、分泌蛋白体外纯化、功能成像分析等技术发现:(1)线虫雄性精液特异蛋白SFP-1(F56D2.8)在交配后行使着调控雌雄同体寿命和传递生殖表型信号的功能,而且这两种调控机制都发生在雌雄同体线虫的肠道细胞内;(2)SFP-1蛋白缺失类NTF2结构域和其C端结构域将会使其失去减少寿命的功能;(3)SFP-1在交配后进入雌雄同体肠道细胞,通过SKN-1通路,促进其表达和入核,进而促进雌雄同体体细胞脂质代谢,导致交配后雌雄同体寿命减短。本项目首次发现雄性精液蛋白对异性的生理调控机制,揭示蛋白跨组织传递生殖信号的作用机制,为重新审视精液蛋白多功能性提供一个重要的范例。
