锌在血管环境中具有适宜的降解速度和良好的生物相容性,近年来成为可降解支架材料的研究热点之一,但目前缺乏材料体系优化和支架植入实验,对锌基支架的在体降解机制、力学退化规律和生物学效应没有深入研究。基于前期优化的高强高塑Zn-Li-Mn合金体系(屈服强度243-369MPa,抗拉强度362-552MPa,延伸率53-103%),本项目将结合腐蚀学和细胞学评价,筛选最优合金成分配比,研究血管环境中的关键化学和力学因素(pH值、氧含量、应变、流体剪切力、脉动压力)对材料和支架降解行为的影响,探明金属离子协同释放与细胞应答之间的关系;通过动物实验,研究锌合金支架在体降解行为与力学退化规律,考察其降解产物对血管修复的组织学影响,揭示Zn-Li-Mn合金支架在血管修复过程中的在体降解机制和生物学效应,为可降解锌基材料在血管环境中的应用提供实验依据和理论基础。

Zinc performs appropriate degradation rates and good biocompatibility in vascular environments, which enables it to become one of the research focuses in biodegradable stent materials. However, very few studies have been done in optimizing zinc-based stent materials. More importantly, the degradation mechanism and mechanical degeneration of zinc-based stents, and their biological effects are unclear due to limited in vivo studies. The Zn-Li-Mn ternary alloys were optimized in our previous study, showing sufficient mechanical properties (yield strength of 243-369 MPa, ultimate tensile strength of 362-552 MPa, elongation to failure of 53-103%) for stent applications. The proposed research aims to optimize zinc alloy composition based on corrosion and cytological evaluations, investigate the influence of key chemical and mechanical factors (pH, oxygen content, strain, fluid shear force, and pulsating pressure) on the degradation behavior of materials and stents, and study the relationship between co-release of metallic ions and cellular responses (endothelial cells, smooth muscle cells, and macrophages). Animal tests will be performed to study the in vivo degradation behavior and mechanical degeneration of the Zn-Li-Mn alloy stent, and histological responses to its degradation products. This project will reveal the in vivo degradation mechanism and biological effects of the Zn-Li-Mn alloy stent during the vascular healing process, and provide the experimental and theoretical foundation for the application of biodegradable zinc-based alloys in vascular environments.
缺血性心脏病是人类头号死因,也是我国疾病主要负担。支架植入是临床治疗缺血性心脏病的主要手段。可降解支架有望在支撑狭窄血管并帮助组织修复后被细胞和组织安全降解、吸收和代谢,从而克服或缓解传统药物洗脱支架(DES)造成的晚期并发症。锌是人体必需微量元素,锌金属在体内显示出良好的可降解性,是除铁、镁外第三种有望用于可降解血管支架研发的医用金属。基于前期设计、优化的Zn-Li-Mn三元高强、高塑锌合金材料体系,本项目主要研究内容包括:综合评测Zn-Li-Mn系合金的力学、腐蚀以及细胞学性能以筛选用于支架制备的最优成分;通过电化学、动态浸泡、在体时序多尺度表征研究Zn-Li-Mn合金支架的降解行为机理;通过组织、细胞学研究Zn-Li-Mn合金支架及其降解产物对血管细胞的生物效应。所取得创新成果如下:(1)Zn-0.8wt.%Li-0.1wt.%Mn合金具有最优的综合性能,抗拉强度552 MPa,延伸率68%,降解均匀性优于纯锌,对血管内皮细胞相容性优异。Zn-0.8wt.%Li-0.1wt.%Mn薄壁支架径向支撑强度150kPa,优于同壁厚Co-Cr合金支架(120kPa);(2)Zn-Li-Mn支架发生降解时,Li元素优先溶出并与体液反应形成碳酸锂等产物,该产物水解引起支架表面局部碱化,进而使得氧化锌、碳酸锌等产物获得热力学稳定性,延缓了支架的降解速率。支架在血流环境中降解速率相对较高、腐蚀形貌均匀,但在内膜环境中降解减缓,降解以局部腐蚀为主。在兔腹主动脉植入6个月内保持良好的力学完整性;(3)Zn-Li-Mn合金裸支架具有一定的抗内膜过度增生作用,6个月时再狭窄率<15%,远低于临床50%标准,与雷帕霉素洗脱Co-Cr合金支架相当。体外细胞结果显示,Zn-Li-Mn合金浸提液对HUVEC内皮细胞相容性良好,0-2倍稀释的Zn-Li-Mn合金浸提液(Zn:1.53-6.13μg/mL;Li:0.25-1μg/mL)可显著促进内皮细胞增殖、抑制平滑肌细胞增殖、迁移。基于以上研究,以第一作者或第一通讯作者在Nat.Comm.、Adv.Mater.、Adv.Health.Mater.等发表论文4篇,申请中国发明专利1项,培养研究生2人。
