蝙蝠是SARS等多种病毒的自然宿主但却不发病,不易患肿瘤,耐受多种体外寄生虫,较同等体型哺乳动物的寿命长十余倍,这些现象都与蝙蝠特殊的免疫系统有关。但目前对蝙蝠免疫系统的适应性进化研究还很少,对以上特殊现象的免疫系统分子调控机制不够清楚。本项目拟对分属两个蝙蝠亚目6科7种蝙蝠的免疫系统做广泛而深入的研究。将具有不同冬眠习性和食性的蝙蝠做对比,研究5种重要免疫器官基因表达谱在同种蝙蝠冬眠和非冬眠期的差异,以及不同食性、不同物种之间的差异,并对重要的免疫相关基因做适应性分子进化分析。我们还将高通量测序所获得的蝙蝠数据与包括人、小鼠的已知哺乳动物数据做比较,在蝙蝠和小鼠中做基因表达试验验证,以发现不同哺乳动物免疫系统的差异。本项目将发现与蝙蝠免疫相关的重要基因及其特殊的分子进化模式,揭示冬眠、食性、物种对蝙蝠免疫器官基因表达谱的影响,为探究蝙蝠较其它哺乳动物更具抗病力和更长寿的分子机制迈进一步。
英文摘要
Bats are natural hosts for many viruses, such as SARS, but do not get sick. They are resistant to tumors and tolerate a variety of ectoparasites. They live more than ten times longer than mammals of the same size. These phenomena are related to the special immune system of bats. However, there is little research on the adaptive evolution of bat immune system, and the molecular regulatory mechanisms of this special immune system remain unclear. This project plans to study the immune system of 7 species of bats belonging to 6 families of the two suborders, and to find the differences between bats with and without hibernation abilities, and bats of different diets. We plan to study the differences of gene expression profiling of 5 important immune organs in the same species of bats during hibernation and non-hibernation periods, and different species of bats with different diets. We will also perform the molecular analysis of adaption evolution of important immune-associated genes. Moreover, we will compare the data of bats obtained from the high throughput sequencing with known data of other mammalian species, including human and mouse, and will perform gene expression experiments in bats and mice, which will find the differences of the immune system in mammals. This project will discover important genes related to bat immunity and their special molecular evolution, reveal the effects of hibernation, diet, population on the gene expression profiles of immune organs in bats, and explore the molecular mechanisms of anti-disease ability and longevity of bats a forward step.
本项目在全国范围蝙蝠主要分布区域,结合本研究团队历年来积累的采样调查的数据,进行蝙蝠采样和调查。从蝙蝠重要的多种免疫器官基因表达谱入手,筛选重要免疫相关基因。对马铁菊头蝠不同状态下的脾脏、淋巴结和骨髓做了转录组测序和分析,在显著性差异表达基因中,筛选出了蝙蝠冬眠期与免疫相关的显著高表达和低表达基因。这些基因是潜在的与蝙蝠抗病毒等病原微生物能力有关的重要分子。.在项目开展过程中,与中国医学科学院病原生物学研究所、中国疾病预防控制中心病毒病所和中国科学院微生物所等多家单位同时对蝙蝠物种携带的冠状病毒进行分析,探讨了病毒的遗传重组、宿主范围、地理分布及跨种传播特征,丰富了对蝙蝠冠状病毒的认知。对2016-2021年以南方为重点的14个省703个蝙蝠栖息地采集的54个蝠种13,064份蝙蝠样本开展组合筛查,在获得142株SARS相关病毒和4株重组病毒后,系统阐述了中国境内蝙蝠携带Sarbecovirus亚属冠状病毒的遗传多样性、宿主特征和变异规律;证实蝙蝠等野生动物宿主的病毒跨物种传播对人类和家畜健康构成了显著威胁,积极监控蝙蝠及其他犬科动物中的病毒性感染,发现、鉴定对人兽健康构成潜在威胁的新冠状病毒并研究其进化变异特征与感染特性,对于防控新发人兽共患传染病、建设国家生物安全体系具有重要意义。对来自中国14个省的13,064份蝙蝠样本进行了冠通过对中国南方代表性蝙蝠物种携带的冠状病毒进行全景式分析,研究鉴定了覆盖10个已知亚属及一个未分类α冠状病毒亚属的1,141个冠状病毒,获得399条全基因组序列,并识别出11个新的冠状病毒进化谱系,揭示了其高度多样性和分布的广泛性。同时,丰富了对蝙蝠冠状病毒的认知,为SARS-CoV-2的溯源及高风险病毒的预警提供了重要数据和指向性线索,有助于了解高致病性冠状病毒从蝙蝠向人类社会的溢出途径和机制,同时也能及时预警具有潜在跨种传播风险的蝙蝠冠状病毒。
