炎症性肠病(IBD)是一种复杂的慢性肠道炎症疾病。研究表明,肠道菌群紊乱是IBD的重要诱发因素,因此,通过靶向调节肠道菌群紊乱开发新型抗IBD药物具有重要意义。本实验室前期研究发现,条浒苔来源的硫酸多糖(ECP)可显著改变小鼠肠道菌群结构,增加肠道乳酸杆菌、双歧杆菌和阿克曼氏菌等益生菌的含量。然而,其能否通过改善肠道菌群紊乱发挥抗IBD作用尚未得到研究。本课题拟从肠道菌群角度出发,围绕这一新型药物作用靶点,以葡聚糖硫酸钠(DSS)诱导的IBD小鼠为模型,结合基因组学、生物信息学和代谢组学技术,尤其是粪菌移植及厌氧微生物体外模拟发酵技术,系统研究ECP的抗IBD作用并阐明其药理学机制,为抗IBD新药的研发奠定理论基础。

Inflammatory bowel disease (IBD) is a complex and chronic inflammatory disease in the large intestine. Accumulating evidence indicates that gut dysbiosis is a major contributor to the development of IBD. In this regard, it is critically important to develop new anti-IBD drugs by targeting the gut microbiota and attenuating dysbiosis. Our previous work in the lab illustrates that a sulfated polysaccharide from Enteromorpha clathrate (ECP) could significantly change the structure of the gut microbiota and increase the abundance of Lactobacillus spp., Bifidobacterium spp. and Akkermansia muciniphila in mice. However, whether or not ECP could help to alleviate IBD by attenuating gut dysbiosis has not been studied. In this project, starting from the perspective of gut microbiota, a new drug target, and applying a dextran sulfate sodium (DSS)-induced mouse model of IBD, we will systematically investigate the therapeutic effect of ECP on IBD and demonstrate its pharmacological mechanisms by combining a serious of techniques including genomics, bioinformatics, metabolomics, and especially fecal microbiota transplantation and in vitro simulated anaerobic fermentation. We hope that our project will lay a foundation for the development of new anti-IBD drugs.
浒苔硫酸多糖(ECP)是绿藻来源硫酸多糖的典型代表,近年来受到广泛关注。研究表明,ECP具有抗氧化、抗凝、抗病毒、抗菌、降血脂及免疫调节活性。但是,其是否具有抗炎症性肠病(IBD)作用并没有研究报道。针对上述科学问题,本课题以葡聚糖硫酸钠(DSS)诱导的IBD小鼠为模型,系统研究了ECP的抗IBD作用。研究表明,ECP口服可以显著缓解DSS引起的IBD。16S rRNA高通量测序表明,ECP可以改善IBD引起的肠道菌群失调,提示ECP可以通过靶向肠道菌群发挥抗IBD作用。基于此,为了进一步鉴定人体肠道中能够介导ECP发挥抗IBD作用的靶标菌株,我们结合自主开发的厌氧微生物培养组学技术构建了人体肠道菌株库。该菌株库共含有119种,累计2052株不同的人体肠道细菌。以该菌株库为基础,我们系统筛选了能够与ECP代谢互作的人体肠道细菌。结合体外发酵和体内动物实验,我们进一步锁定人体肠道中的Parabacteroides distasonis是能够介导ECP发挥抗IBD作用的关键靶标细菌。研究表明,P. distasonis不仅可以发酵利用ECP产生对宿主有益的乙酸和丙酸,同时其口服也可以显著缓解IBD小鼠的体重降低、结直肠缩短、便稀以及肠道出血等炎症反应。依托上述菌株库,我们进一步筛选获得了Bacteroides xylanisolvens、Bacteroides salyersiae和Bacteroides uniformis等三种具有显著抗IBD作用的新型肠道益生菌。此外,通过与临床医生合作,结合体外模拟发酵、16S rRNA高通量测序及生物信息学分析技术,我们进一步研究了ECP对IBD患者肠道菌群的调节作用。研究表明,ECP可以改善IBD患者的肠道菌群失调,促进其肠道中具有抗炎作用益生菌的生长。此外,考虑到海洋糖类资源的丰富性和多样性,我们同时比较研究了其他糖类分子的抗IBD作用。通过研究,我们发现了另外一类具有显著抗IBD作用的糖类分子低聚古罗糖醛酸(PG)。与ECP类似,PG同样可以通过靶向肠道菌群发挥抗IBD作用。上述研究为ECP及其他海洋糖类抗IBD药物的开发奠定了理论依据。依托本项目,负责人指导毕业硕士生1名,共同指导毕业博士生2名,授权中国发明专利1项,以通讯作者发表SCI论文9篇,其中中科院1区Top论文3篇。
