青少年抑郁症严重危害青少年身心健康,其机制不清。研究报道外泌体与主要精神疾病相关,但其是否与青少年抑郁相关尚不清楚。我们前期发现外泌体可损伤海马神经元,导致青幼期小鼠出现抑郁行为;转录组测序发现青少年抑郁患者外泌体lncRNA DLX6-AS1异常表达,并与临床特征相关;数据库预测提示DLX6-AS1与ERBB4均可结合miR-146b-5p,现有研究报道miR-146b-5p和ERBB4均与抑郁有关;预实验进一步发现miR-146b-5p可抑制靶基因ERBB4表达,该效应能被DLX6-AS1所削弱。因此推测外泌体DLX6-AS1进入海马神经元结合miR-146b-5p,进而调控ERBB4表达引起海马神经元损伤,导致青少年抑郁。本研究拟证明外泌体信号传递在青少年抑郁中的作用,阐明DLX6-AS1介导海马神经元损伤导致青少年抑郁的机制。为青少年抑郁症脑分子机制研究及临床诊疗提供新途径和靶点。

Depression in adolescents can have a serious impact on the physical and mental health of individuals yet its pathogenesis remains to be fully elucidated. Studies have shown that exosomes are associated with major mental disorders yet the role of exosomes in adolescent depression is unclear. Our initial studies showed that exosomes can damage hippocampal neurons and cause depressive behavior in young mice that closely resembling the clinical symptoms in humans. Transcriptome sequencing showed the abnormal expression of exosomal lncRNA DLX6-AS1 in adolescent patients with depression and these changes correlated with the clinical characteristics of patients. Database predictions showed that both DLX6-AS1 and ERBB4 could bind to miR-146b-5p. However, existing studies have reported that both miR-146b-5p and ERBB4 are related to depression. Preliminary experimental showed that miR-146b-5p could inhibit the expression of the target gene ERBB4 and that this effect could be weakened by DLX6-AS1. Therefore, we predict that exosomal DLX6-AS1 enters the hippocampal neurons and binds miR-146b-5p to regulate the expression of the target gene, ERBB4. ERBB4 damages hippocampal neurons and promotes adolescent depression. This study aimed to demonstrate the role of exosomal signaling in adolescent depression and to clarify the mechanism of DLX6-AS1-mediated hippocampal neuron injury that causes adolescent depression. Our results provide new approaches and identify novel targets to study the molecular mechanisms and clinical treatments of depression in adolescent patients.
青少年抑郁症(major depressive disorder,MDD)严重危害青少年群体的健康成长和生命安全,其发病机制不清。细胞外囊泡(extracellular vesicles,EVs)是细胞间信息传递的载体,携带蛋白、脂质和RNA等分子,其中miRNAs是EVs的重要组成部分,在大脑中具有多种重要作用。研究发现,EV-miRNAs可能在精神疾病的发病机制中具有重要作用,但其在青少年MDD发病中的作用及机制仍不清楚。本研究从临床和动物两个层面开展工作,核心内容和重要成果如下:. 在临床研究中,我们设计了横断面和前瞻性研究,采用高通量测序和RT-PCR法检测血清EV-miRNAs表达水平,并评估受试者的临床特征、童年创伤和人格特征。研究结果发现:1)血清EV-miRNAs的表达失调与青少年MDD密切相关,基于血清EV-miR-450a-2-3p和情感虐待组成的模型可以辅助诊断青少年MDD;2)血清EV-miRNAs表达水平与抗抑郁治疗反应不相关,人格特征的外向性与抗抑郁治疗反应相关。基于人格特征组成的模型可以有效预测青少年MDD的抗抑郁治疗反应。. 血清EVs的来源非常复杂,其作用机制很难确定。因此,在动物研究中我们采用L1CAM抗体的免疫捕获法,从血清EVs中富集神经源性EVs(即L1CAM+ EVs)。我们将青春期正常小鼠随机分组,尾静脉移植不同来源的血清L1CAM+ EVs,并对小鼠进行行为学测试、海马Nissl染色、mRNA全基因组测序。研究结果发现:青少年MDD来源的血清L1CAM+ EVs可以诱导青春期小鼠出现抑郁样行为和海马神经元损伤,miR-375-3p是血清L1CAM+ EVs诱导青春期抑郁发病的潜在驱动因子,miR-375-3p可能通过Cadm2、Cacna2d1和Casz1的蛋白互作效应调控了以MAPK信号通路为核心的复杂网络分子,从而导致了青春期抑郁的发生。. 通过上述研究,我们为青少年MDD提供了潜在的辅助诊断工具和治疗反应预测模型。更重要的是,我们的发现为理解青少年MDD病理机制提供了新的视角,并可能为开发有效的青少年MDD诊断和治疗模式提供新靶点和途径。
