缺氧缺血(HI)引起的神经元死亡是新生儿缺氧缺血性脑病(HIE)的重要病理基础。申请人前期研究报道丹参酮Ⅰ(TsI)有神经元保护作用,但机制不明。近期发表文章发现HI 后PTEN核转移增加与神经元死亡有关,上调Aagab抑制PTEN核转移。预实验又发现TsI可上调 Aagab、减少HI后PTEN核转移、降低SESN2及p-JUN蛋白水平、抑制自噬激活。据此推测:TsI发挥神经元保护作用的可能分子机制是通过上调Aagab减少PTEN核转移抑制p-JUN-SESN2/AMPK通路活化抑制自噬激活。本项目拟探讨:①TsI是否通过抑制自噬激活发挥神经元保护作用,②PTEN核转移是否通过p-JUN-SESN2/AMPK通路激活自噬导致神经元死亡;③TsI是否通过上调 Aagab减少PTEN核转移抑制HI后神经元死亡。旨在阐明TsI发挥神经元保护作用的分子机制,为治疗HIE提供新的靶点和思路。

Neuron death caused by hypoxia ischemia (HI) is the important pathological basis of neonatal hypoxic-ischemic encephalopathy (HIE). Our previous study has reported that tanshinone I (TsI) exists neuroprotective effect. However, the underling mechanisms are poorly understood. Recent study also found that the neuron death induced by HI was related to the increase of PTEN nuclear translocation, and Aagab up-regulation suppressed PTEN nuclear translocation. In addition, our pre-experiment data showed that TsI treatment can up-regulate Aagab, reduce PTEN nuclear translocation induced by HI, decrease SESN2 and p-JUN protein level, and suppress the activation of autophagy. Therefore, we speculate that the possible mechanism of the neuroprotective effect of TsI is to inhibit the activation of autophagy through inhibiting p-JUN-SESN2/AMPK pathway by up-regulating Aagab and decreasing PTEN nuclear translocation. The present project intends to explore: ① TsI exerts neuroprotective effect whether through inhibiting autophagy. ② The neuron death caused by PTEN nuclear translocation whether through activating autophagy by activating p-JUN-SESN2/AMPK pathway. ③ TsI suppress the death of neuron induced by HI whether through up-regulating Aagab to reduce PTEN nuclear translocation. The present study aimed to clarify the molecular mechanisms of the neuroprotective effect of TsI. Knowledge gained from this study may provide new target and clinical therapeutic methods for HIE.
由围产期窒息引起的新生儿缺氧缺血性脑病(HIE)是导致新生儿死亡和儿童持久残疾的主要因素之一。在发达国家,HIE的发病率约为每1,000名活产婴儿1至4例,在发展中国家高达每1000名活产婴儿~26例,其中25%患儿死亡,而35%的幸存者留有长期神经系统后遗症。由于目前针对HIE没有效的治疗方法HIE患儿预后很差,给家庭和社会带来了沉重的负担。因此,寻找神经保护疗法对于指导临床治疗HIE至关重要。本项目中我们的研究发现p-JUN-SESN2-AMPK通路在体内和体外缺氧缺血(HI)模型中都被激活。下调JUN减少新生缺氧缺血性脑损伤(HIBD)大鼠的神经元丢失,改善肌力、运动平衡及协调功能以及空间学习和记忆障碍。同时我们发现通过多肽Tat-K13(一种靶向PTEN K13残基的短干扰肽)抑制PTEN核转移抑制HI引起的p-JUN-SESN2-AMPK通路的激活。我们还发现HI后自噬被激活,Tat-K13阻断PTEN核转移通过抑制p-JUN-SESN2-AMPK通路抑制自噬。此外,我们发现Tat-K13阻断PTEN核转移通过抑制自噬减少HI导致的神经元丢失和改善行为缺陷。最后我们还发现TsI通过上调Aagab抑制p-JUN-SESN2-AMPK通路从而抑制自噬激活进而发挥神经保护作用、改善HI诱导的行为损伤。这些结果表明,HI后PTEN核转移通过激活p-JUN-SESN2-AMPK通路激活自噬损伤神经元,而TsI/Tat-K13通过抑制p-JUN-SESN2-AMPK通路抑制自噬促进神经功能恢复,从而改善HIBD大鼠行为损伤,这可能为新生儿HIE的临床治疗提供新的治疗方法。
