肿瘤微环境在癌症发生发展中发挥着非常关键的作用,其中肿瘤相关成纤维细胞(CAFs)是肿瘤微环境中含量最多的细胞之一,参与建立并重塑细胞外基质,并通过分泌多种细胞因子参与肿瘤-基质细胞的共同进化,是肿瘤侵袭转移发生的“驱动者”之一。因此靶向抑制CAFs的动员和招募将会成为肿瘤治疗的新策略。我们在前期实验中发现ESCC细胞通过分泌FGF2去动员FGFR2+纤维祖细胞增殖,继而招募入肿瘤组织中分化成CAFs,但CAFs的招募和分化分子机制尚不清楚。因此,本课题将寻找招募FGFR2+纤维祖细胞的关键趋化因子;应用全转录组测序及生物信息学技术揭示CAFs分化的分子机制;最后通过中和抗体阻断CAFs的动员和招募,确定其在肿瘤治疗的可行性。本项目研究将拓展我们对CAFs起源、动员、招募及分化的认识,为从源头上靶向抑制CAFs产生继而抑制肿瘤生成提供理论基础,具有重要的科学理论价值及临床指导价值。

Tumor microenvironment (TME) plays a critical role in cancer development and progression. Cancer-associated fibroblasts (CAFs) are one of the most infiltrated stroma cells in TME. Functionally, CAFs build up and remodel the extracellular matrix, accelerate tumor-stroma co-evolution via releasing numerous regulatory factors. Besides, CAFs are a main driver of the invasion-metastasis cascade in many cancer subtypes. Therefore, it is conceivable that mobilization and recruitment of CAFs might be new potential targets in cancer therapy. We previously found that FGFR2+ fibrocyte progenitors in bone marrow could be mobilized by FGF2 secreted from cancer cells. Moreover, they could be recruited into tumor tissue and differentiate into functional CAFs. However, the precise molecular mechanisms of CAFs recruitment and differentiation remain unclear. In this proposal, we plan to find the key chemokine responsible for recruiting FGFR2+ fibrocyte by in vitro and in vivo experiments. In addition, the molecular mechanisms underlying their differentiation will be addressed via high-throughput RNA-sequencing and bioinformatics analysis. Finally, the therapeutic potential of neutralizing antibodies blocking the process of CAFs mobilization and recruitment will be evaluated in mouse model. Our aim is to provide more insight for better understanding the origin, mobilization, recruitment and differentiation of CAFs, and to optimize CAFs targeting strategy for more effective cancer treatment.
肿瘤微环境在癌症的发生和发展中发挥着至关重要的作用。其中,肿瘤相关成纤维细胞(CAFs)是肿瘤微环境中通过分泌多种细胞因子来参与肿瘤细胞与基质细胞的共目进化。来源于肿瘤相关成纤维细胞(Cancer-associated fibroblasts, CAFs)的分泌蛋白在食管鳞状细胞癌(Esophageal squamous cell carcinoma, ESCC)发生发展过程中起着至关重要的作用。在我们之前的研究结果中,我们发现来自FGFR2+造血干细胞的纤维细胞可以被ESCC细胞动员、招募到肿瘤区域中,然后分化为CAFs。在本项研究中,我们发现肿癌细胞可以通过分泌FGF2动员骨髓内FGFR2阳性的前体细胞分裂并进入外周血成为FGFR2阳性循环纤维前体细胞,肿癌细胞再通过分泌CXCL12与FGFR2阳性循环纤维前体细胞膜上的CXCR4相互作用,将它们招募到肿瘤组织中,进而分化成FGFR2阳性CAFS。我们发现在动物模型中可以利用抗体(anti-CXCR4)通过阻断CXCL12/CXCR4结合,抑制食管癌的生长。.进一步研究找到了2个CAFs分泌的蛋白MFGE8和LTBP2,系统研究了它们在食管癌发生发展中的作用。结果发现显示MFGE8+CAFs的数量随 ESCC 进展而增加。TCGA-ESCC数据集的相关性分析显示在ESCC中,MFGE8或LTBP2的高表达与总生存率差(P<0.05),无病生存率差(P<0.05)密切相关。功能实验表明 MFGE8 可能是 CAF 重塑肿瘤环境的重要因子,它通过与整合素结合 αVβ3/αVβ5 受体,激活 PI3K/AKT 和 ERK/AKT 通路。从而促进肿瘤的生长和转移。另一项研究发现LTBP2 与整合素 α6β4 相互作用,导致 Src 信号通路激活。使用针对 LTBP2 的拮抗抗体可增强 ESCC 细胞对化疗的敏感性,表明 LTBP2 是提高 ESCC 治疗效果的有前景的治疗靶点。.动物体内研究证明用抗体阻断MFGE8或LTBP2都能显著抑制肿瘤体内生长。.最后我们还利用单细胞测序技术研究了4例临床食管癌病人,样本包括原发灶、淋巴结转移灶和癌旁组织,共分析85 263 个单细胞转录组,结果发现POSTN+CAFs与淋巴结转移灶微环境的建立密切相关。这些发现对控制食管癌淋巴结转移提供了新的治疗策略。
