基于患者诱导性多能干细胞(iPS)培养的类脑器官能够在体外模拟人脑的发育过程和结构特点,是研究多种脑疾病的理想模型。申请人的研究关注2019年发表的一项大规模遗传学研究鉴别出了一个新的阿尔茨海默病(AD)风险基因ADAMTS1。在前期研究中已发现ADAMTS1与APP蛋白存在相互作用,抑制Aβ42的产生。对AD患者和健康对照的测序鉴别出一个可以影响该基因表达的启动子区域SNP与AD相关。本研究拟利用申请人建立的类脑器官技术,在iPS分化获得的类脑器官中明确(1)能否通过干预ADAMTS1来减少患者神经细胞Aβ产生;(2)阐明ADAMTS1参与APP切割的分子机制。本研究将有助于理解风险基因ADAMTS1在AD等神经退行性疾病中的作用机制,并为AD的病因学研究及新治疗靶点研发提供参考。

Alzheimer's disease (AD) is one of the most common serious neurodegenerative diseases. the postulated hypothesis that the accumulation of Aβ peptides initiates the pathogenesis of AD has received widespread attention. In recent years, the drug targets identified solely from mouse models lack the potential to be converted into clinical applications, which suggests that we must decipher the origin of pathogenesis and molecular targets based on findings from human studies. A large population-based genome-wide association study published in 2019 identified a new AD-associated gene ADAMTS1, but the function of this gene in the nervous system is unclear. Our previous study found that the metalloproteinase ADAMTS1 interacts with APP protein (APP produces Aβ). APP can be cleaved into a previously unknown fragment by ADAMTS1 overexpression (we name it sAPPats1), and the generation of Aβ42 was inhibited.Cerebral organoids derived from patient specific-iPS by three-dimensional culture may be an ideal disease model of AD. In this study, organoids from AD patients with TTT mutations in the promoter region ofADAMTS1, and organoids with modified gene mutation by CRISPR-Cas9 will be studied to determine: (1) whether AD can be treated by interfering with ADAMTS1 expression level or molecular function, (2) what is the molecular mechanism of ADAMTS1 involved in APP hydrolytic cleavage. This study will contribute to understand the mechanism of risk gene ADAMTS1 in neurodegenerative disease such as AD, and provide a reference for the research of etiological mechanism of AD and the development of new therapeutic targets.
阿尔茨海默病(AD)是目前发病率最高的神经退行性疾病,2019年与2021年两项大型全基因组关联研究显示ADAMTS1基因与AD风险相关,但对于该基因在AD中的作用机制不清。因此本研究在分子和细胞生物学层面,探究ADAMTS1在脑内尤其是海马的细胞特异性分布,以及神经活动性和复杂的认知活动对ADAMTS1的表达调控,并探究ADAMTS1对APP的水解功能及分子相互作用机制;最后在动物模型层面,利用5×FAD小鼠构建ADAMTS1过表达模型,对ADAMTS1水解APP的功能进行体内验证。与此同时,AD的发病机制至今仍没有研究清楚,其中遗传因素占重要地位,PSEN1基因作为家族性AD最高风险致病基因被广泛关注。. 基于患者诱导性多能干细胞(iPS)培养的人脑类器官能够在体外模拟人脑的发育过程和结构特点,是研究多种脑疾病的理想模型。利用AD患者iPS系、CRISPR-Cas9基因编辑、脑类器官诱导体系等技术平台,分析新发现的遗传风险位点ADAMTS1对AD的保护性机制,验证PSEN1基因的E363Q突变和IVS2c.-54+2T>A突变对细胞造成的作用影响,有助于揭示更多风险基因遗传突变在神经退行性疾病中的作用机制。. 通过本项目的实施,我们首先利用细胞模型验证了ADAMTS1会影响APP水解通路,此后发现高水平ADAMTS1可以显著延缓5×FAD小鼠的认知衰退,并在模型小鼠的海马组织中通过检测Aβ相关蛋白的表达情况进一步明确了ADAMTS1对认知改善的分子机制。同时,利用AD患者自体iPS来源的人脑类器官,配合CRISPR/Cas9基因编辑技术,发现同样是APP水解酶的PSEN1基因的E363Q和IVS2c.-54+2T>A这两个突变,均具有致病性。研究成果为AD等脑疾病的发病机制研究及新治疗靶点研发提供了参考价值。
