近年来,抗生素抗性基因(antibiotic resistance genes,ARGs)对人类健康的潜在风险持续增加。河岸过滤系统是水力交互、物质代谢及能量传递的关键区域,也是环境微生物抗性产生的“热点”区域及潜在抗性基因库。因此,探索ARGs在河岸过滤带原位微生物群落中的生态响应机制及传播行为,具有重要的科学意义。该项目拟利用河岸过滤室内模拟装置及原位观测系统,借助高通量测序及宏基因组学技术,分析不同介质赋存生境下典型ARGs在微生物群落中的富集、传播规律,观测微生物群落结构及代谢功能变化,探讨微生物群落对抗生素及ARGs的生态反馈机制;进一步地,通过投加携带目标ARGs的载体微生物,利用q-PCR及FISH技术,研究河岸过滤系统不同介质特征及水力停留时间下ARGs及载体微生物的截留效应,构建其丰度预测模型并评估潜在风险,以期为河岸过滤技术及饮用水源安全风险防控提供科学依据。

In recent years, the potential risks of antibiotic resistance genes (ARGs) on human health have been widely discussed. Riverbank filtration (RBF) system is a key area of hydraulic interaction, material metabolism, and energy transfer. It is also a "hot spot" for environmental microbial resistance inducing and a resistance gene bank. Therefore, it is of great scientific significance to explore the mechanism of the transfer and migration of ARGs in the RBF system, as well as the ecological response mechanism of the in-situ microbial community. In this research, RBF in-situ sites and simulation systems are established and high-throughput sequencing techniques and metagenomics analysis method are used to identify typical ARGs and host microorganisms, whose transfer and accumulation mechanism in the microbial community, as well as the influencing factors, are also analyzed. The feedback mechanism and ecological effect of the in-situ microbial community to antibiotics along with their resistance are also discussed by monitoring the community structure and metabolic function variation in the RBF system. Furthermore, by adding selected ARGs together with its host microorganism into the RBF system and adopting q-PCR\FISH technique, the migration pattern and the interception effect of the ARGs in the RBF system with different porous medium and typical hydraulic residence time are studied. A prediction model for ARGs and hosts can be established to evaluate the potential risk. This research can provide a scientific reference to the application of RBF technology and drinking water source security risk prevention and control.
近年来,抗生素抗性基因(antibiotic resistance genes,ARGs)对人类健康的潜在风险持续增加。河岸过滤系统是水力交互、物质代谢及能量传递的关键区域,也是微生物抗性产生的“热点”区域及潜在抗性基因库。因此,探索ARGs在河岸过滤带的传播行为及其微生物群落的生态响应机制具有重要的科学意义。.本研究选取淮河流域地表水饮用水源地、沙颍河子流域、里下河泻湖相沉积区及长江三角洲冲洪积沉积区作为典型研究区域,并利用河岸过滤室内模拟系统,借助高通量测序、qPCR技术、宏基因组测序及Metacompare模型等,进行了研究区域原位生境地表水及其潜流带抗性背景特征调查,识别了地表水及其潜流带ARGs传播机制、评估了其水源安全风险;分析了不同介质赋存生境下磺胺类ARGs在微生物群落中的富集、传播规律,观测了微生物群落结构及代谢功能变化,探讨了微生物群落对抗生素及 ARGs的生态反馈机制;进一步地,通过投加携带目标ARGs的载体微生物,研究了河岸过滤系统不同介质特征及水力停留时间下ARGs及载体微生物的截留效应。.通过上述研究,(1)系统地揭示了饮用水源地、山溪性河流-地下水、泻湖相、冲洪积相区域河流-地下水交互带的地表水、地下水及其潜流带沉积环境抗生素、ARGs、微生物群落及MGEs的赋存及其共存网络特征,识别了野外原位生境ARGs的传播机制,评估了水源安全风险;(2)基于野外生境中识别出的关键影响因素,开展了室内批实验及柱实验,解析了不同介质、不同水力停留时间及营养水平特征下,外源性磺胺类抗生素及其ARGs在河岸过滤带的迁移规律与累积效应,提出了河岸过滤带去除磺胺类抗生素及其抗性基因的水力停留时间及其介质条件的参数优化。研究成果丰富了对河流-地下水交互带ARGs环境化学行为的认识,同时,也为河岸过滤技术及饮用水源安全风险防控提供了科学依据。
