The brown planthopper (BPH), Nilaparvata lugens (Stål) (Hemiptera: Delphacidae) is a typical resurgent pest induced by pesticides. Previous studies showed that BPH feeding on rice plants sprayed with jinggangmycin (JGM) resulted in changes in biochemical substances and enhancing fat metabolism in BPH female, stimulating BPH reproduction. However, its regulatory mechanism remains unclear. The recent studies showed that the glucose and sucrose contents was significantly increased in the rice plants sprayed with jinggangmycin (JGM), a fungicide widely used in China for controlling rice sheath blight, Rhizoctonia solani, compared to control rice plants, resulting in the up-expression of insulin-like peptide 2 (ILP2) and activation of the target of rapamycin (TOR) signaling pathway in BPH female after feeding on rice plants sprayed with JGM. Based on the reported roles of ILPs in perception and regulation of sugar nutrition in Drosophila melanogaster and other insects, we proposed the scientific hypothesis that BPH ILP2 in response to JGM-induced increase in sugar content of rice plants regulates reproductive growth by activing the TOR signaling transduction pathway. Based on our previous accumulated achievements, this project aims to test this hypothesis using RNAi interference, bioassay, western blot and immunofluorescence approaches. Firstly, we intend to unravel the mechanism of ILP2 in BPH female triggers the TOR signaling pathway in response to JGM-induced increase sugar content of rice plants; secondly, we want to reveal the mechanism of the ILP2/TOR signaling pathway regulates BPH reproductive growth in response to JGM-induced increase sugar content in rice plants. By comprehensive analysis of results derived from this project, the molecular mechanism of JGM stimulated BPH fecundity from the perspective of BPH insulin sensing changes in sugar nutrition of will be illustrated. The results will provide new ideas for the development of new pesticide targets and resistance management in BPH.
褐飞虱是一种典型的药剂诱导的再猖獗型水稻重大害虫。以前研究表明,褐飞虱取食JGM喷雾处理的水稻后其体内生化物质改变和脂肪代谢增强,刺激生殖。但其作用机制不清楚。最近研究发现JGM喷雾处理诱导水稻体内蔗糖和葡萄糖含量增加,导致取食该处理的水稻后激活褐飞虱雌虫体内ILP2/TOR信号通路转导。因此,我们提出“褐飞虱ILP2响应JGM诱导水稻糖含量增加激活TOR信号通路刺激生殖”的科学假设。我们的研究结果表明,与对照相比,JGM处理的分糵期水稻体内的葡萄糖和蔗糖含量升高,水稻光合作用增加,SPS、INV2和INV3基因表达上调。褐飞虱取食JGM处理的水稻、外源注射葡萄糖和胰岛素的褐飞虱雌虫体内ILP2/TOR信号通路关键基因转录水平与蛋白水平显著上调,Foxo基因表达下降,JH合成酶基因JHAMT及其受体Met基因表达水平显著上调和JH滴度含量增加,Vg表达水平也相应上调。外源注射二甲双胍(Metformin,降血糖药)、LY294002和Rapamycin后褐飞虱体内ILP2/TOR信号通路关键基因表达下降,Foxo基因表达上调,JH合成酶基因JHAMT及其受体Met表达下降和JH滴度含量下降,Vg表达水平也下降。与dsGFP对照相比,外源JHA类似物注射dsILP2处理的褐飞虱雌虫后则出现相反的结果。JGM+dsILP2处理的褐飞虱雌虫产卵量与JGM处理相比下降约63%,子代种群数量下降约58%,与JGM+dsGFP处理相比雌虫产卵量下降约61%,子代种群数量下降了约54%。半田间试验也出现类似的结果。该结果阐明了ILP2感知高葡萄糖信号激活TOR信号通路调控褐飞虱生殖的作用机制,从胰岛素感知营养因子变化角度揭示了JGM刺激褐飞虱生殖的分子机制。该结果也将为褐飞虱开发新的农药靶标及抗性治理提供新的思路,为田间合理用药和褐飞虱的综合治理提供新的指导策略。
