微生物降解是去除环境类固醇激素污染的主要途径;然而国内外对微生物降解雌激素的代谢途径及机制的研究很少。通过国家自然科学基金项目的持续支持,申请者一直开展海洋来源的特定微生物降解有机污染物的机制研究;最近的研究发现微生物菌群之间可通过独特网络结构彼此联系、相互协助,在污染物降解中发挥重要的作用。本项目拟以兼具污染物“源”和“汇”特点、并具有丰富微生物资源的红树林湿地为研究对象,利用稳定同位素示踪和16S rRNA基因高通量测序联用的技术,分析典型红树林中参与降解雌二醇的微生物核心菌群以及雌二醇的中间代谢产物;利用宏基因组学、宏转录组学等技术,研究降解雌二醇的关键功能基因;进一步借助细胞内融合基因技术(epicPCR),构建红树林微生物核心菌群协同降解雌二醇途径。预期成果有助于揭示红树林环境中雌激素的降解机制、并明确发挥作用的功能菌,从而为红树林污染环境修复提供理论支撑。

Microorganisms play an important role in removing the pollution of steroid hormone in environment. However, less studies were focused on the pathway and mechanisms of estrogen degradation. The applicant has been working on the mechanism of organic pollutants degradation (polycyclic aromatic hydrocarbons and estrogen) by the microorganisms from marine environment, which was supported by a series of NSFC projects. Recently, it was found that microbial communities can contact and assist with each other through a unique network structure to degrade pollutants, indicating microbial communities harbor an important function in pollutants degradation. In this study, the microorganisms will be collected from mangrove wetland, which is considered as the “source” and “sink" of persistent organic pollutants and contains abundant microbial resources. First, we will perform the enrichment culture of the microorganisms by using 13C labeled estradiol as substrate to detect the intermediate metabolites of estradiol degradation and analyze the core microbial communities involved in the estradiol degradation. Moreover, the key functional genes in estradiol degradation were identified by using meta-genomics combined with meta-transcriptome. Furthermore, by using the epicPCR (Emulsion, Paired Isolation and Concatenation PCR), the pathway of synergetic degradation of estradiol by core microorganism communities from mangrove will be constructed. The expected results will be helpful to reveal the mechanism of estrogen degradation in mangrove environment and identify the functional microorganisms involved in the degradation, thus this study can provide theoretical support for the bioremediation of environmental pollutes in mangrove.
日益严重的雌激素污染对生态环境和人类健康造成严重危害。红树林湿地作为污染物的“源”和“汇”,蕴藏着丰富微生物资源,这些微生物对环境中雌激素的去除有着至关重要的作用。本项目以红树林沉积物微生物菌群为研究对象,探究微生物菌群降解雌二醇(E2)的机制。利用E2为唯一碳源对红树林菌群富集培养,获得6个高效降解E2的菌群。这些菌群可将E2转化为雌酮(E1)、4-羟基雌酮(4-OH-E1)等中间产物,并最终以E2为唯一碳源生长。基于16S rRNA基因高通量测序,明确了E2降解的核心菌群为Oleiagrimonas、Pseudomonas、Terrimonas和Nitratireductor;同时,利用宏基因组和宏转录组明晰了E2降解的功能基因,包括17β-羟基类固醇脱氢酶(17β-HSD)、雌酮4-羟化酶等;进而通过功能基因异源表达验证了E2降解起始步骤关键酶基因gcaD、BH5_1、YX23_1和17β-HSD(HN14)等,并鉴定了后续降解的关键基因fmo3、BH5_2和YX23_2。最后,结合产物检测与“多组学”手段重构了红树林富集菌群降解E2的途径。此外,通过分子生物学及生物信息学手段,成功将降解功能基因溯源定位于富集菌群的纯培养菌株,并通过菌株复配手段解析了细菌之间通过底物交叉互养协同降解E2的机制。
