随着淡水水体富营养化的日益加剧,饮用水及水产品中微囊藻毒素(MCs)的污染问题也日益突显。MCs有多达几十种亚型,且其毒性差异非常大。因此,仅检测MCs总量不能准确评估样品的真实毒性。然而,目前还没有合适的方法能够用于MCs主要亚型的现场快速分析。为了解决这一问题,本项目提出了采用阿达玛变换拉曼光谱结合表面增强拉曼(SERS)的解决方案,拟实现以下三个目标:1)拟研制一台适合产业化的阿达玛变换拉曼光谱仪样机,解决传统色散型便携式拉曼光谱仪灵敏度相对较低、仪器抗震防潮性能差的问题;2)研发一系列高选择性、高灵敏度的SERS检测基底,用于饮用水、复杂环境水体及水产品中MCs主要亚型的快速检测;3)拟通过阿达玛变换拉曼光谱仪的引入、提高SERS基底的均匀性及采用内标比率的方法,解决SERS定量方法重现性差的普遍难题。本项目的顺利实施,将为精准评估饮用水及水产品中MCs毒性提供一种强有力的手段。
英文摘要
As the eutrophication of water is increasing globally, the pollution of microcystins (MCs) in drinking water and aquatic products is becoming more and more prominent. MCs have as many as dozens of subtypes and their toxicity varies greatly. Therefore, detection of the total amount of MCs cannot accurately assess the true toxicity of the sample. However, hitherto, there are still no suitable instruments and methods that can be used for the field rapid analysis of MCs subtypes. To solve this problem, this project proposes a solution using Hadamard transform Raman spectrometer combined with surface enhanced Raman scattering (SERS) technology. The following three objectives are to be achieved based on the proposed approach: 1) A novel Hadamard transform Raman spectrometer is expected to be produced. Comparing with the traditional dispersive portable Raman spectrometer, the proposed Hadamard transform Raman spectrometer should have better sensitivity and anti-seismic and moisture-proof performances; 2) A series of high selectivity and high sensitivity SERS active substrates for rapid detection of MCs subtypes in drinking water, complex environmental water bodies and aquatic products are developed; 3) It is proposed to solve the general problem of poor reproducibility of SERS quantitative methods by introducing Hadamard transform Raman spectrometer, improving uniformity of SERS substrate and adopting internal standard. The proposed project will provide a powerful means for accurate assessment of MCs toxicity in drinking water and aquatic products.
