酒蒸黄精可增强其补气养阴作用,但其炮制机理尚未阐明。现有的黄精酒蒸机理认为多糖是药效成分。我们前期研究发现黄精中还有多种非糖类成分在酒蒸后含量上升或下降,故黄精酒蒸机理研究还需从以下方面完善:(1)黄精酒蒸后补气养阴作用增强原因除了还原糖含量上升外,皂苷、黄酮等成分的变化也是因素之一。(2)也可能是某些成分在酒蒸作用下发生了包括Millard反应在内的一些化学反应导致质变或量变,并伴有酒蒸后体内代谢产物发生变化而实现的。拟开展工作如下:(1)通过动物实验(整体、器官和分子水平)对黄精酒蒸前后补气养阴作用进行比较研究;(2)采用谱效分析等手段探讨酒黄精药效物质与效应的相关性,通过UPLC-Q-TOF-MS辨识补气养阴物质基础;(3)酒蒸后饮片中含量升高和体内吸收增加的单体的药效学研究。从体内外化学成分差异与药效变化相关性角度揭示黄精酒蒸增效的机理,为黄精质量控制、炮制工艺规范化提供科学依据。

Wine steamed Polygonati Rhizoma can enhance invigorating Qi and nourishing Yin effect, but the processing mechanism has not been clarified. The existing steamed mechanism of Polygonati Rhizoma considers that polysaccharide is its medicinal ingredient. Our previous studies found that there are a variety of non-sugar components whose contents go up or down after steamed in Polygonati Rhizoma, therefore, the mechanism study of wine steamed Polygonati Rhizoma needs to be improved from the following aspects: (1) the invigorating Qi and nourishing Yin effect of wine steamed Polygonati Rhizoma is enhanced because the increase of reducing sugar content, also the change of saponins, flavonoids and other components is one of the factors. (2) it could also be some ingredients occur a number of chemical reactions under the action of wine steaming, including the Millard reaction, which result in qualitative or quantitative changes, and accompanied the changes of the metabolites in body after wine steamed. The proposed research work is as follows: (1) the invigorating Qi and nourishing Yin effects of Polygonati Rhizoma before and after steaming are studied by animal experiment (whole body, organ and molecular level); (2) spectral and effect analysis is used to study the correlation between the pharmacodynamic substances and the effect of wined Polygonati Rhizoma, identificate the material basis of invigorating Qi and nourishing Yin by UPLC-Q-TOF-MS; (3) the pharmacodynamics study of monomer compounds with increased content in decoction pieces and increased absorption in vivo after steamed. Thus from the correlation between the change of chemical constituents in vitro and in vivo and the change of pharmacologic action, to reveal the mechanism of synergistic effect of wine steamed Polygonati Rhizoma, provide scientific basis for the quality control of Polygonati Rhizoma, the standardization of processing technology.
本项目为国家自然科学基金青年基金,执行期3年。本项目以黄精和酒黄精为研究对象,在总结黄精研究文献的基础上,系统开展了黄精和酒黄精补气养阴作用的比较,从一般状态观察、体重和摄食量变化、脏器指数、病理组织学评价、血常规检查、生化指标、免疫组化、免疫荧光、PCR和WB等方面证实酒黄精补脾气养阴作用优于生黄精。开展了黄精酒蒸后糖类和非糖类成分变化研究,发现黄精酒蒸过程中多糖和寡糖分解生成单糖(果糖、葡萄糖和木糖),使多糖和寡糖减少,单糖含量增加;黄精酒蒸后还原糖含量大幅度增加;黄精酒蒸后多糖分子量增大,多糖的单糖组成种类不变,但所组成单糖的含量发生了变化;黄精酒蒸后产生了新成分5-HMF、黄精碱A、DDMP,并且这3种成分含量显著增加,考虑可通过测定这3种成分的含量有效区分黄精和酒黄精。开展了黄精和酒黄精挥发性和非挥发性成分差异研究,发现1-丙醇、2-庚酮、2-正戊基呋喃、1-己醇、乙酸甲酯、1-戊烯-3-醇、3-甲基丁醇等可作为黄精的特征挥发性物质,1,8-桉树脑等可作为酒黄精的特征挥发性物质;黄精和酒黄精的差异代谢产物有22个,生黄精中氨基酸类成分质量分数高,炮制品中单糖的质量分数高,可能因为在加热酒蒸过程中,多糖水解为单糖和寡糖等小分子糖类成分,虽然随着反应的进行,小分子糖类和氨基酸类成分发生美拉德反应,使两者含量减少,但总的来说,小分子糖类还是增加的,因为小分子糖类容易被人体吸收,故炮制品的补益作用增强,另外,酒黄精中酒渣碱的质量分数高于生品,提示其可能更有利于提高免疫作用。该研究初步阐明黄精酒蒸增效机理,为黄精炮制工艺规范化和饮片的质量控制,黄精饮片临床合理应用提供科学依据。中药炮制影响黄精临床疗效的研究应纳入复方中进行深入研究(如黄精丸、二精丸等),以体现中医临床用药的特点,同时阐明复方中药物炮制增效的机制。
