Avian reovirus (ARV) is an important pathogenic agent endangering poultry industry. The ARV infection is mainly characterized by clinical symptoms such as arthritis/tenosynovitis and retarded development syndrome. Previous research results show that ARV infection induced apoptosis in host cells, leading to tissue damage in infected chickens. However, the molecular mechanism underlying ARV-induced apoptosis is still not very clear. Our data from previous studies indicate that ARV infection altered the expression profiles of host miRNAs, in which the expressions of mir-29a-3p and mir-200a-3p were significantly up-regulated, and overexpression of both miRNAs suppressed ARV-induced apoptosis. Thus, based on these findings, this project will be conducted to investigate into the molecular mechanisms underlying ARV-regulated expressions of miR-29a-3p and miR-200a-3p in host cells and suppression of apoptosis by the miRNAs using the tools in Molecular Biology, Molecular Virology and Immunology, to elucidate the molecular basis of ARV-mediated effect on miRNA expression and to unravel the molecular mechanism underlying suppression of apoptosis by ARV infection and its effect on virus replication. The results from this study will provide theoretical basis to uncover the pathogenesis of ARV-host interaction and help to formulate effective control measures for ARV infection.
禽呼肠孤病毒(Avian reovirus, ARV)是严重危害国内外家禽养殖业的重要病原,其感染可引发病毒性关节炎、腱鞘炎和矮小综合征等临床症状,并可引起免疫抑制,现有疫苗存在免疫应答不足及安全性方面的局限性。本研究聚焦ARV与宿主互作机制,系统解析了宿主miRNA在调控病毒致病过程中的关键作用,为新型防控策略的开发提供了理论依据。本研究通过高通量测序发现,ARV感染DF-1细胞后显著上调宿主miR-29a-3p和miR-200a-3p表达。进一步功能研究表明,过表达miR-29a-3p和miR-200a-3p能显著抑制ARV诱导的细胞凋亡,而敲低内源性miRNA则增强细胞凋亡。病毒滴度检测结果显示,miRNA过表达可降低细胞培养物中ARV滴度,沉默miRNA则促进病毒增殖,表明二者通过调控凋亡通路影响病毒复制效率。此外,靶基因验证实验显示,miR-29a-3p直接靶向凋亡执行蛋白Caspase-3,能够抑制其蛋白表达。敲低miR-29a-3p后Caspase-3表达增加,而Caspase-3抑制剂处理可降低病毒滴度,说明miR-29a-3p通过拮抗Caspase-3介导的凋亡,抑制病毒复制。miR-200a-3p则靶向调控生长因子受体结合蛋白2(GRB2),敲低miR-200a-3p可增强ARV诱导的细胞凋亡,并促进 GRB2 表达。抑制内源性GRB2的表达则会降低细胞培养物中的病毒滴度。这表明miR-200a-3p通过靶向 GRB2抑制ARV感染引起的细胞凋亡和病毒复制。在此基础上,研究进一步发现ARV感染会上调宿主miR-30c-5p的表达。miR-30c-5p可以通过靶向自噬基因ATG5抑制LC3-I向LC3-II转化,从而阻断病毒自噬依赖性复制。充分阐明病毒感染影响宿主miRNAs表达的分子基础以及miRNAs抑制细胞凋亡的分子机理与生物学意义,在RNA水平上解析了ARV的致病机理,系统揭示了宿主miRNA通过靶向凋亡核心分子抵抗ARV的感染的分子机制,为开发基于RNA调控的新型疫苗及诊断技术奠定了重要理论基础,并对提升ARV综合防控能力具有显著科学价值与应用潜力。
