糖尿病与癌症二者之间存在的高风险相关性已被流行病学证实,但糖尿病诱发肿瘤的机理仍然不明。本项目前期研究结果表明糖尿病病理因素糖基化终末产物(AGEs)可诱发肝细胞中心体扩增,而中心体扩增是导致基因组不稳定性的重要因素,这是细胞癌变的前提。因此探究AGEs诱发中心体扩增的分子机理是揭示糖尿病诱发癌症的关键。转录组分析表明AGEs显著上调肝细胞PLK1与CEP20的表达,siRNA干扰证实PLK1和CEP20是AEGs诱发肝细胞中心体扩增的关键蛋白。中心体扩增的原因最终均可归结为复制循环的紊乱和中心体组装的异常。因此本课题将聚焦PLK1-CEP20调控中心体复制周期及组装的机理研究,揭示PLK1-CEP20表达或功能异常导致中心体扩增的分子机理。并验证PLK1-CEP20表达或功能异常导致的中心体扩增是否是AGEs诱发肝细胞发生恶性转变的主要因素。这将为探究糖尿病诱发肿瘤提供理论基础。

The high-risk correlation between diabetes and cancer has been confirmed by epidemiology, but the mechanism of diabetes-induced tumors is still unknown. Preliminary research results of this project indicate that AGEs, a pathological factor of diabetes, can induce hepatocyte centrosome amplification which could lead to genome instability, a prerequisite for cell carcinogenesis. Therefore, exploring the molecular mechanism of centrosome amplification induced by AGEs is the key to revealing that diabetes induces cancer. Transcriptome analysis showed that AGEs significantly up-regulated the expression of PLK1 and CEP20 in hepatocytes, and siRNA interference confirmed that PLK1 and CEP20 are key proteins for AEGs to induce centrosome amplification. The cause of centrosome amplification can ultimately be attributed to the disorder of the replication cycle and the abnormal assembly. Therefore, this research will focus to reveal the molecular mechanism of how PLK1-CEP20 regulating the centeosome duplication cycle and assembly, and verify centrosome amplification caused by the disorder expression of PLK1-CEP20 is the main reason for hepatoma genesis induced by AGEs. This will provide a theoretical basis for exploring diabetes-induced tumors.
在本项目的资助下,主要探究了糖尿病病理因素糖基化终末产物(AGEs)与中心体扩增以及肝细胞恶性转变之间的关系。首先通过收集糖尿病及肝癌患者临床数据及肿瘤样本,经临床数据分析发现糖化血红蛋白能够显著上调肝癌的患病风险,且细胞模型实验表明AGEs的处理能够促进永生化肝细胞的恶性转变。通过病理切片以及细胞模型实验检测发现AGEs能够显著上调肝细胞中心体扩增水平,且中心体扩增可促进肝细胞克隆形成、增殖、侵袭迁移能力。抑制中心体扩增可抑制AGEs诱发的肝细胞克隆形成、增殖、转移等恶性肿瘤细胞特征的发展。表明中心体扩增在介导AGEs诱发肝细胞恶性转变过程中发挥重要作用。接下来通过转录组测序,以及Crispr-cas9基因敲除和过表达等基因表达调控工具,通过逆转基因表达水平发现EGR1、DNMT3B、PLK1、CEP20、CCDC116因子在AGEs诱导肝细胞中心体扩增调控中发挥重要作用。EGR1作为转录因子结合并激活PLK1基因表达,DNMT3B通过甲基化修饰CEP20启动子抑制CEP20表达。AGEs处理上调EGR1,下调DNMT3B,因此促进PLK1和CEP20转录表达。PLK1可磷酸化修饰CEP20,进而抑制CEP20泛素化降解,提高CEP20稳定性,并促进其与CCDC116结合。CEP20与CCDC116的结合促进了CEP20中心体定位。免疫荧光标记中心体及中心粒特异性蛋白,实验检测结果表明过量的CEP20在中心体上的聚集会导致中心粒过度复制,进而导致中心体扩增。因此通过本研究我们证实了糖尿病病理因素AGEs与肝癌发生之间的关系,并揭示了AGEs通过EGR1/DNMT3B-PLK1-CEP20-CCDC116信号轴诱发中心体扩增促进肝细胞恶性转变的分子机制,这可能为糖尿病及癌症的相关治疗提供新的思路或靶点。
