Excellent blood compatibility is required for the successful application of blood-contacting materials. Unfortunately, the healthy endothelium is the only known surface that is blood compatible in all respects. Natural blood vessels have unique multi-scale micro-nano composite structures. In addition, the abundant glycosaminoglycans (GAGs) on the surface of vascular endothelium can ultimately protect the blood vessels and keep the blood flowing. Inspired by the chemical and topographical features of the vascular endothelium, we aim to develop poly(dimethylsiloxane) (PDMS) materials with improved hemocompatibility by combining the chemical composition and surface topography of GAG mimetics in a biomimetic way. A facile method to produce bromine-containing PDMS surface (PDMS-Br) with topographical features (surface patterning and micro-/nanoscale surface topographies) will be developed by a one-step multicomponent thermocuring procedure and replica molding. Different GAG mimetics will be introduced on the flat and topographical PDMS-Br surface by visible light-induced graft polymerization. In addition, air plasma assisting microcontact deprinting and printing transfer approaches will be used to fabricate gold film patterns and PDMS patterns. Then differernt GAG mimetics may be introduced in the areas of gold film patterns and PDMS patterns, respectively, to form hetergenerous patterned surfaces modified with GAG mimetics. The influence of chemical compisition and surface topography of GAG mimetics on the hemocopatibility of the modified surfaces will be investigated. The possible mechanism will also be discussed. A biomimetic principle that combining chemical composition and surface topography of GAG mimetics may provide a new strategy to design blood compatible materials.
本项目利用糖胺聚糖(GAG)类似物的化学组成和拓扑结构的协同效应,成功制备了一系列具有优异血液相容性的高分子材料。首先,通过同步结合多组分热固化工艺和复制模塑法,制备了具有有序图案结构和微纳米复合结构的含溴引发剂的聚二甲基硅氧烷(PDMS)表面(PDMS-Br)。然后,通过可见光诱导接枝聚合法,在具有不同拓扑结构的PDMS-Br表面引入GAG类似物。同时,通过图案化金膜转印法和金-硫键自组装技术,制备了具有均质和异质化学组成的GAG类似物改性表面。研究了GAG类似物化学组成和拓扑结构双重因素对材料表面血液相容性的影响规律及其作用机制。进一步地,以上述影响规律为指导,制备了一系列基于GAG类似物的多功能血液接触材料。通过本项目的研究,较好地完成了预期研究计划,达到了预期研究目标。针对本项目的研究,目前一共发表学术论文14篇,其中影响因子大于5的论文11篇,中科院Top期刊论文7篇,其中2篇文章(Regenerative Biomaterials, 2024, 11, rbae096; Biomacromolecules, 2024, 25, 7202)被邀请作为当期杂志的正封面文章。这些发表的论文被国内外学术刊物引用近100次,表明本项目的研究成果得到国内外学者的关注。项目的研究成果申请并获得国家发明专利2篇;项目负责人参与撰写了科学出版社出版的书籍1部,培养和协助指导硕士生12名;项目成员积极参与国内外学术会议交流,累计参与交流7次、合计15人次。GAG类似物化学结构单元和拓扑结构的有机结合为制备血液相容性材料提供了新方法和新思路,且该策略能方便地拓展到其他生物医用高分子材料上,有望为制备具有优异血液相容性的生物医用材料提供独特的解决方案。
