果实在贮藏和运输过程中容易失水,造成严重的品质劣变和采后损失,这不仅与环境条件如温度、湿度和空气流速有关,而且与果实的角质层结构和成分紧密相连。果实成熟过程中角质层结构和成分不断变化,是特定基因时空表达的综合结果,受到转录因子的调控。目前关于果实角质层的研究主要集中于角质层结构的形成和成分分析,转录调控的研究鲜有报道。课题组前期研究发现NAC转录因子NOR-like1参与果实角质层发育,与野生型对照果实相比,NOR-like1基因编辑果实容易失水,且角质层发育缺陷,但是具体的分子机制尚不明细。本项目拟利用野生型,NOR-like1基因编辑和超表达果实为材料,通过细胞结构观察,角质层代谢成分分析,RNA-seq和ChiP-seq技术等挖掘NOR-like1调控的角质层代谢靶基因,解析NOR-like1调控角质层形的分子机制。研究结果将为将来培育耐失水的果实品种提供理论依据。

Fruit is easy to lose water during storage and transportation, resulting in serious quality deterioration and postharvest loss, which is not only related to environmental conditions such as temperature, humidity and air flow rate, but also closely related to the structure and composition of fruit cuticle. The changes of structure and composition of fruit cuticle are the result of the expression of specific genes in time and space during its ripening, which are regulated by transcription factors. At present, the research on fruit cuticle is mainly focused on the formation of cuticle structure and component analysis, and its transcription regulation is rarely reported. Our previous study found that the NAC transcription factor NOR-like1 participated in the development of fruit cuticle. Compared with the wild-type control fruit, the NOR-like1 gene editing fruit is easy to lose water, and its fruit cuticle development is defective, but its molecular mechanism is not clear. This project will use wild-type, NOR-like1 gene editing and overexpression fruit as materials, through cell structure observation, metabolic component analysis, RNA-seq and ChIP-seq technology, etc. to dig the target gene of NOR-like1 which involved cuticle formation, and establish the transcriptional regulation mechanism of NOR-like1 regulating cuticle metabolize . The results will provide a theoretical basis for the cultivation of water-loss-resistant fruit varieties in the future.
果实在贮藏和运输过程中容易失水,造成严重的品质劣变和采后损失,这不仅与环境条件如温度、湿度和空气流速有关,而且与果实的角质层结构和成分紧密相连。目前关于果实角质层的研究主要集中于角质层结构的形成和成分分析,转录调控的研究鲜有报道。课题组前期研究发现NAC转录因子NOR-like1参与果实角质层发育,与野生型(WT)对照果实相比,SlNOR-like1基因编辑果实容易失水,且角质层发育缺陷,但是具体的分子机制尚不明细。本项目主要研究内容包括WT和SlNOR-like1基因编辑果实的采后持水能力分析;WT和SlNOR-like1基因编辑果实的角质层结构表征;WT和SlNOR-like1基因编辑果实角质层的化学成分检测;WT和SlNOR-like1基因编辑果实外果皮的转录组学分析;SlNOR-like1下游角质层代谢相关靶基因鉴定。项目通过表型观察、角质层结构和化学成分表征,结合转录组学分析和靶基因验证,证明了SlNOR-like1通过靶向激活角质沉积关键基因SlGPAT6和SlCD2正调控角质沉积,通过靶向抑制蜡质生物合成与转运相关基因SlKCS1、SlCER1-2、SlWAX2和SlLTPG1-like负调控蜡质积累。项目解析了SlNOR-like1调控番茄果实角质层形成的分子机制,完成了项目的研究目标。另外还研究了SlNOR-like1在番茄花粉发育、番茄果实大小以及番茄果实软化方面的功能。项目研究结果对于利用SlNOR-like1改善园艺产品采后失水、产量和软化具有参考价值。
