中性粒细胞缺陷相关疾病临床危害大、治疗效果不佳。研究中性粒细胞发育及机制,从源头上改善中性粒细胞缺陷,具有重大临床意义。斑马鱼具有与人类高度相似的基因组、造血发育过程,是研究造血发育重要的动物模型。申请人前期创新性发现TEAD1a可从髓系最早期开始促进斑马鱼中性粒细胞发育,通过CRISPR/Cas9诱导tead1a基因突变可导致斑马鱼中性粒细胞缺陷表型,而过表达TEAD1a及下游分子通路可挽救缺陷表型,深入研究发现这种表型很可能是通过下游gata1/notch通路介导。据此,本项目拟进一步深入研究,阐明TEAD1a在斑马鱼中首先通过抑制gata1转录促进髓系前体细胞(CMP)向粒系-单核系原始细胞(GMP)分化,之后通过上调notch信号通路进一步促进中性粒细胞分化发育的机制。本项目将为阐释中性粒细胞发育提供新的理论依据,为中性粒细胞缺陷的治疗提供新策略。
英文摘要
The diseases related to neutrophil deficiency can do great harm to patients and present poor response to treatment. It is of great importance to study the mechanism of neutrophil development and fundamentally cure its defect. Zebrafish has a high similarity of genome and hematopoietic development process with human, and therefore an ideal model for hematopoietic development study. Previously, the applicant found that TEAD1a can promote neutrophil development in zebrafish from the earliest stage of myelopoiesis. Mutation of tead1a induced by CRISPR/Cas9 led to defective neutrophil development in zebrafish, and overexpression of TEAD1a or its downstream molecular pathways rescued the defective phenotype. Furthermore, we found that this phenotype was probably mediated by downstream gata1/notch pathway. Based on this, this project intends to verify that TEAD1a firstly promotes the differentiation from CMP to GMP by inhibiting the transcription of gata1, and then promotes neutrophil differentiation and development by promoting the notch signaling pathway. This project will provide a new theoretical basis for the interpretation of neutrophil development and a new strategy for the treatment of neutrophil deficiency.
中性粒细胞减少症是一种中性粒细胞缺陷相关的疾病,临床风险大、预后不良,目前集落刺激因子(G-CSF)作为其主要的治疗方案,所产生的中性粒细胞却存在缺陷,如发育异常、功能钝化等,导致疗效不佳,亟待更加有效的中性粒细胞刺激因子填补目前的治疗空缺。在本研究中,我们首先通过数据库分析总结出比G-CSF更高阶的髓系发育潜在刺激因子群,通过应用其基因表达序列进行大规模斑马鱼胚胎显微注射,筛选出中性粒细胞刺激作用最明显的转录因子TEAD1a,随后通过构建斑马鱼中性粒细胞缺陷模式生物模型,证实了TEAD1a或TEAD1a/YAP结合蛋白缺陷,可导致髓系分化严重受损,最终导致中性粒细胞减少症的表型,这一发现提示TEAD1a及TEAD1a/YAP相互作用在髓系细胞分化中的关键作用。我们随后通过逐步的机制探究、测序分析和表型验证,发现TEAD1a或TEAD1a/YAP1相互作用首先通过直接抑制gata1启动子促进髓-红系前体细胞向髓系前体细胞分化,再通过Notch信号通路促进髓系前体细胞激活,而最终促进中性粒细胞造血分化及功能成熟的下游关键因子为spi1和cebpa,即TEAD/YAP-notch-spi1/cebpa信号通路的失能与中性粒细胞减少症的发生发展密切相关。不仅如此,我们的研究进一步发现TEAD1a或TEAD/YAP相互作用对中性粒细胞造血的影响始于造血过程的极早期阶段(10-18hpf),远在G-CSF发挥作用前,且并未明显依赖于G-CSF发挥作用,这一发现不仅揭示了造血过程中的极早期阶段激活中性粒细胞发育的新机制,加深了我们对髓系细胞分化起始时间窗口的认识,为理解中性粒细胞减少症的发病机制提供了新的视角,而且极大得拓宽了中性粒细胞减少症的新治疗靶点探索,对于提高中性粒细胞减少症患者的治疗效果和改善预后具有重要的临床应用价值。
