猪脂肪性状与肉品质和饲料转化率密切相关,脂肪生成机制研究具有重要科学意义。野生型p53诱导磷酸酶1(Wip1)是重要调控因子,其对免疫和繁殖的调节作用研究较多,但在调控小鼠脂肪生成中的重要功能刚刚受到关注,且调控猪脂肪性状的功能未见报道。在前一项国家基金资助下,本团队获得了Wip1基因编辑猪,揭示了调控猪繁殖性状的机制,并发现其脂肪沉积明显减少。本项目将利用本团队独有的Wip1基因编辑梅山猪新种群,针对Wip1磷酸酶,采用dDIA磷酸化蛋白质组学新技术,规模化获取脂肪组织差异磷酸化位点及其对应蛋白信息,通过其与Wip1的互作及与猪脂肪表型性状的关联分析,阐明Wip1调控目标蛋白特异位点磷酸化从而影响脂肪生成的机制。本项目以在脂肪沉积方面具有代表性的我国优良地方种质资源为素材,在国内外同行大量前期工作基础上,阐明Wip1调控猪脂肪生成的新机制,为培育优质高效猪新品种提供新的分子基础。

The fat traits of pigs are closely related to meat quality and feed conversion, and the study of fat formation mechanism is of great scientific significance. Wild-type p53-induced phosphatase1 (Wip1) is an important regulatory factor, which has been widely studied in the regulation of immunity and reproduction. Its important function in the regulation of mouse adipogenesis has just been concerned, but its function in the regulation of pig adipogenesis has not been reported. With the support of the former grant of National Natural Science Foundation of China, our team obtained the editing pig of Wip1 gene, revealed the mechanism of regulating the reproductive traits of pigs, and found that the fat deposition of editing pigs was significantly reduced. In this project, by using our team's unique new genetic resources of Wip1 gene editing Meishan pig, and aiming at the fact that Wip1 is phosphatase, we will use direct data independent acquisition(dDIA) phosphorylation proteomics new technology to obtain the information of different phosphorylation sites and their corresponding proteins in fat tissues on a large scale. Through the analysis of interaction with Wip1 and the correlation analysis with the fat phenotype of pigs, we will clarify that the specific phosphorylation of the target proteins regulated by Wip1 and then their regulation mechanism of fat formation. In this project, based on the representative local germplasm resources of our country in terms of fat deposition and a large number of high-level previous work of our colleagues at home and abroad, the new mechanism of regulating fat production of pigs by Wip1 will be clarified, which will provide a new molecular basis for the cultivation of high-quality and efficient new varieties of pigs.
本项目利用基因编辑梅山猪新种群揭示了WIP1基因调控猪脂肪生成的机理。首先系统检测了WIP1基因在不同猪种中各组织的表达,发现了WIP1基因在脂肪和肌肉的表达规律,证明其参与调控猪肌肉和脂肪组织的生长发育。利用体外脂肪细胞分化模型探讨了Wip1在3T3-L1前脂肪细胞增殖分化中的作用。同时获得了野生型梅山猪和WIP1基因敲除梅山猪的前体脂肪细胞,发现WIP1基因敲除通过调控相关分化基因的表达抑制了梅山猪前脂肪细胞的成脂分化能力,并采用转录组和磷酸化蛋白组学方法构建了相关调控网络。此外,本研究对WIP1基因敲除梅山猪和对照梅山猪进行了生长性能和屠宰性能测定,并利用蛋白质组学和磷酸化蛋白组学方法在个体和群体水平上构建了WIP1基因影响猪脂肪生成的调控网络,解析了WIP1基因影响猪脂肪生成的机理。本研究还进行了WIP1基因与猪生长性状及精液品质的关联分析,获得了可作为分子标记的位点。本研究从细胞水平、个体水平和群体水平解析了WIP1对梅山猪脂肪生成的作用及调控机制,可为我国地方猪遗传资源的改良提供基因资源和理论基础。本项目发表文章4篇,其中1篇文章入选2023年度中国农业期刊行业优秀论文;申请国家发明专利1项,并授权;培养研究生3名。
