代谢重塑在细胞命运决定中具有重要作用。近年来发现在小鼠多能胚胎干细胞重编程为全能性二细胞样细胞的过程中细胞代谢也发生了重塑,然而代谢重塑的规律和作用机理仍不清楚。前期工作我们发现转录因子Glis1调控糖酵解代谢重塑促进多能性获得(Nature Metabolism 2020),近期工作发现在多能性维持中Glis1还能使小鼠多能胚胎干细胞重编程为全能性二细胞样细胞。我们推测Glis1在二细胞样细胞获得中具有重要作用。本项目拟在此基础上进行如下研究:1)确定Glis1对二细胞样细胞获得过程的调控作用;2)揭示Glis1在二细胞样细胞获得过程中对代谢的调控;3)利用代谢探针 “SoNar” 等技术阐明Glis1通过代谢重塑促进二细胞样细胞获得的分子机理。本研究将阐明Glis1调控全能性细胞获得的代谢重塑规律,并揭示Glis1促进全能性细胞获得的机理,为探索细胞命运转变的代谢调控提供重要参考。

Metabolic remodeling plays an important role in cell fate determination. In recent years, it has been found that cellular metabolism is also remodeled during the process of mouse pluripotent embryonic stem cells reprogramming into totipotent 2C-like cells, however the patterns and mechanisms of metabolic remodeling are still unclear. We previously found that Glis1 regulated glycolytic metabolic remodeling and promoted pluripotency acquisition (Nature Metabolism 2020). Recently, we found that Glis1 can also reprogram mouse pluripotent embryonic stem cells into totipotency 2C-like cells. We hypothesized that Glis1 plays an important role in totipotency 2C-like cell acquisition. This project intends to further below:1) to confirm the role of Glis1 in totipotency 2C-like cell acquisition; 2) to uncover the regulation of Glis1 on metabolism pathway in totipotency 2C-like cell acquisition; 3) to demonstrate the molecular mechanisms of metabolic remodeling promoting totipotency 2C-like cell acquisition by metabolic probe “SoNar” and other technologys. Our study is expected to clarify the metabolic remodeling pattern during cell totipotency acquisition, and reveal its metabolic regulation pathway and the mechanisms of Glis1 promoting totipotency cell acquisition. Our project would provide an important idea for exploring the metabolic regulation for cell fate determination.
多能干细胞具有无限的增殖、自我更新以及多向分化的潜能,即多能性。研究多能干细胞命运调控不仅能帮助我们更好地理解早期胚胎发育过程,在干细胞再生医学应用方向也具有巨大前景。近年来的研究发现,在体外培养的小鼠多能干细胞中,存在着低比例的一群细胞,这群细胞具有和体内二细胞期胚胎相似的转录组特征以及分化成胚外组织的潜能,被称为全能性二细胞样细胞。同样在人的体外培养的多能干细胞中则存在着全能性八细胞样细胞,这些细胞为研究生命的起始及再生医学提供了非常好的工具。有趣的是,多能性干细胞和全能性干细胞可以互相转变,然而多能干细胞如何转变为全能干细胞的机理仍不清楚。. 本项目中通过分析公开的转录组学数据,项目负责人发现母系转录因子Glis1不仅在正常发育过程中的卵子、合子及二细胞期胚胎高表达,而且在由小鼠多能干细胞重编程得到的全能性二细胞样细胞中也高表达,这提示Glis1在全能性二细胞样细胞获得过程中具有重要作用。进一步,通过建立小鼠全能性二细胞样细胞报告基因细胞系,发现过表达Glis1可以显著提高小鼠多能干细胞重编程为全能性二细胞样细胞的比例。深入研究发现Glis1在Dux基础上极大地促进了全能性二细胞样细胞的产生。机制研究发现,Glis1并不是通过调控糖酵解或线粒体代谢来促进全能性细胞获得,而有可能是通过调控细胞内钙离子浓度相关信号通路来实现。此外,项目负责人利用高精度蛋白质乳酸化修饰组学等技术发现,全能干细胞比多能干细胞具有更高水平的蛋白质乳酸化修饰密度,且蛋白质乳酸化修饰在全能干细胞和多能干细胞中具有不同的基序,提示在这两类细胞中具有不同的蛋白质乳酸化修饰调控机制。. 本项目的研究将帮助我们深入理解哺乳动物胚胎早期发育过程及多能干细胞命运转变过程,具有重要的理论意义与研究价值。
