英文摘要
The most common metastatic sites of colorectal cancer are liver and lung, which is the leading cause of death in colon cancer patients. CXCL12/CXCR4 activation is associated with liver metastasis of colorectal cancer, while CXCR7, another receptor of CXCL12, has recently been found to be related to lung metastasis of colorectal cancer, but the mechanism is unclear. From the perspective of tumor microenvironment, this project aims to explore the new mechanism of CXCL12/CXCR7 biased signaling transduction promoting the activation of cancer-associated fibroblasts (CAFs) in the tumor microenvironment through exosome-encapsulated miRNAs and thereby promoting lung metastasis of colorectal cancer. Taking high-throughput miRNAs sequencing as the starting point, we elucidated the molecular mechanism of CXCR7-overexpressing colorectal cancer cells promoting the activation of CAFs by upregulating exosomal miR-155-5p and miR-146a-5p using clinical tumor samples, animal models and cell lines. More importantly, we first proposed that CXCL12/CXCR7 biased signal activation promoted YAP1 nuclear translocation by recruiting β-arrestin 1, through which exosomal miRNAs were upregulated. These results reveal the messenger effect of exosomal miRNAs in promoting tumor metastasis, and provide new targets and theoretical evidence for inhibiting organotropic lung metastasis of colorectal cancer and precision medicine-guided treatments.
转移是结直肠癌恶性发展的最终结局,常发生肝转移和肺转移,成为临床治疗的痛点问题。近年来发现肿瘤细胞与微环境基质细胞的相互作用在器官特异性转移过程中起着重要作用,而外泌体成为介导细胞间通讯的重要信使。我们通过对临床结肠癌病人标本和Villin-CXCR7转基因小鼠炎症性结肠癌模型研究,发现CXCR7表达与肿瘤相关成纤维细胞(CAFs)激活标志物之间存在显著的正相关。RNA测序显示CXCR7高表达结肠癌细胞及其外泌体中miR-146a-5p和miR-155-5p水平的均呈现显著增加。本项目从临床结肠癌组织样本、小鼠动物模型、细胞分子水平系统研究,取得以下创新性成果:(1)发现CXCR7高表达结肠癌细胞通过激活微环境CAFs促进肺转移;(2)发现CXCR7高表达结肠癌细胞外泌体中miR155-5p、miR-146a-5p通过调控靶基因促进CAFs激活,为以外泌体为载体的miRNAs药物研发提供靶点;(3)发现CXCL12/CXCR7偏向信号通过招募β-arrestin 1促进YAP1入核上调miRNAs表达,为靶向CXCR7偏向受体抑制剂研发提供理论依据;(4)发现结肠癌病人血清外泌体中miR155-5p、miR-146a-5p水平与肿瘤转移和预后相关,为诊断和预测结肠癌病人转移提供血清标志物。本研究不但从肿瘤和微环境相互作用的角度,阐明了趋化因子受体CXCR7促进结肠癌肺转移的新机制,揭示外泌体miRNAs(miR-155-5p、miR-146a-5p)以及YAP1核转位是药物干预的潜在靶点,有助于开发以外泌体为载体的miR-155-5p、miR-146a-5p抑制剂药物抑制结肠癌转移。结肠癌病人血清外泌体中miR155-5p、miR-146a-5p表达水平成为诊断和预测结肠癌病人转移的重要血清标志物。我们的研究为抑制结肠癌转移提供新的思路和治疗策略。
