如何促进新生细胞分化成熟为有功能的神经元是成体神经干细胞(NSCs)修复颅脑创伤(TBI)的关键问题。过去认为Treg细胞通过免疫抑制机制发挥作用;新近研究发现Treg细胞还可影响干细胞和组织再生修复,但Treg细胞对TBI后成体神经再生的作用尚不清楚。本课题以挖掘Treg细胞独立于免疫抑制之外的促再生新机制为出发点,在前期一系列研究结论的基础上,调控Treg细胞浸润动力学使其高峰期与分化成熟关键期相吻合;在临床病例分析基础上,利用TBI模型和ere转基因小鼠探讨Treg细胞基于Amphiregulin/EGFR机制直接促进新生神经元功能成熟的作用;并利用体外细胞和脑片培养的非免疫微环境,进一步证实促再生作用并不依赖于免疫抑制。本课题以TBI病理进程原有的免疫细胞亚群为靶点,探寻可借助促再生直接作用和免疫抑制间接作用的综合调控措施,将有助于实现成体干细胞研究在TBI的临床转化。

How to promote newborn cells differentiating into mature and funtional neurons is the key problem in adult neural stem cells (NSCs) to repair the neurological function after traumatic brain injury (TBI). It has been agreed that the role of T cell subgroup Treg cells is mainly through the mechanism of immunosuppression. Recent studies have found that Treg cells can also affect the stem cells and tissues repair. However, the effect and mechanism of Treg cells on the regeneration of adult neurogenesis after TBI are still elusive. The purpose of this study is to explore a new mechanism for promoting regeneration of Treg cells independent of immunosuppression. The peak of infiltration kinetics of Treg cell is regulated to coincide with the critical stage of differentiation and maturation which has confirmed on the basis of previous research conclusions. Based on the analysis of clinical cases, TBI model and ere transgenic mice are used to study the role of Treg cells in directly promoting the functional maturation of newborn neurons through mechanism of Amphiregulin/EGFR pathway. Meanwhile, the nonimmune microenvironment of cells and brain slices culture in vitro is used to further demonstrate that the effect of regeneration is not dependent on the immunosuppression. In this study, the immune cell subgroup in TBI pathology is adopted as the regulatory targets, and a comprehensive measure by both directly promoting regeneration and indirect immunosuppression is established, which is more conducive to the clinical transformation of adult stem cells research in TBI.
背景:创伤性脑损伤因其具有极高的死亡率与致残率,给社会和家庭带来了沉重的负担。TBI 后神经功能重建一直是困扰研究学者和临床医生的难题。关于成体神经干细胞(NSCs)的探索与研究,很大程度冲击了传统中枢神经系统不可再生的传统观点。如何有效利用TBI 诱导成体NSCs 大量增殖的现象和规律,改善TBI 后的认知功能,甚至改善TBI 后的运动、感觉功能,己经成为干细胞治疗TBI 研究领域关注的热点问题。TBI 后诱导的NSCs 分化成熟为有功能的神经元,是成体NSCs 发挥修复作用的首要前提。然而,新生细胞分化成熟的能力低下严重阻碍了其对TBI 神经功能的修复。研究发现调节性T 细胞因其免疫抑制作用成为损伤后修复的热点,但关于其独立于免疫抑制而直接促进神经再生的研究尚少。.内容:本研究首先在动物实验中通过定向敲除海马齿状回区(DG)的Treg 细胞,探究Treg 细胞耗竭对TBI 后神经再生和神经修复的影响;再通过体外补充AREG重组蛋白的方式,检测海马齿状回区神经再生及小鼠认知功能的变化情况,明确Treg细胞耗竭是否通过AREG影响TBI后的神经再生与损伤修复,AREG是否可以逆转因Treg 细胞缺失造成的神经损伤。体外实验通过在NSCs培养体系中加入AREG重组蛋白,检测NSCs增殖和分化能力的改变,探究免疫抑制作用之外AREG对神经干细胞增殖分化的影响。.结果:(1)动物实验通过流式细胞术分析了TBI后Treg细胞的时间动力学规律;EdU标记实验结果表明TBI后Treg细胞缺失会导致细胞增殖减少,DCX免疫荧光染色结果表明TBI后Treg细胞缺失会抑制NSCs分化能力;而通过体外补充AREG重组蛋白后,NSCs 的增殖与分化能力较之前得到恢复;Morris 水迷宫实验结果表明TBI 后Treg 细胞缺失导致小鼠认知功能退化,而体外补充AREG重组蛋白后小鼠认知功能得到恢复。(2)细胞实验证明AREG可以独立于Treg细胞的免疫抑制作用之外调控NSCs 增殖,诱导NSC分化。
