英文摘要
Conjugated linoleic acid(CLA)is a new type of functional oil. Lactic acid bacteria have become the research hotspot because of their ability to specifically synthesize the active isomers of cis-9, trans-11-CLA and trans-10, cis-12-CLA isomers with stronger probiotic functions. Changing the external environment and the expression of CLA genes can regulate CLA synthesis, but the regulation mechanism is not clear. The applicant previously found that LttR transcriptionally regulates CLA biosynthesis, and ArgR2 is a potential regulator in CLA synthesis. Based on this, this project takes Lactobacillus plantarum AR195 as the research object, further explores other regulators affecting CLA biosynthesis by DNA affinity chromatography and LC-MS/MS. Then constructs the mutant strains, and analyzes the regulation of regulators on the target gene from the metabolic and transcriptional levels. The specific binding sites will be located by DNase I footprinting method. A cooperative regulatory network of CLA metabolic transcriptional regulators will be constructed by RNA-seq and molecular interaction data analysis. The regulatory mechanism of CLA biosynthesis will be studied at the molecular level. Through the implementation of this project, it will deepen the understanding of CLA biosynthesis of lactic acid bacteria and lay a theoretical foundation for the design of metabolic engineering of lactic acid bacteria to increase the yield of CLA.
共轭亚油酸(CLA)是一种具有重要药理作用和营养价值的功能性脂肪酸,已被广泛研究并应用于食品、保健品和临床领域。乳酸菌因能高效合成c9,t11-CLA和t10,c12-CLA活性异构体成为当前研究热点。近年来的研究表明,改变外部环境条件或调控CLA相关基因的表达,均能够影响CLA的合成过程。然而,具体的转录调控机制仍未被充分揭示。本研究以植物乳杆菌AR195为研究对象,重点探讨了其CLA生物合成的转录调控机制。在前期工作基础上,通过全基因组组学分析,获得了该菌株的基因组信息,并识别了CLA生物合成相关基因的启动子区域。通过DNA亲和层析技术,我们筛选出了多个与CLA生物合成相关的潜在转录调控因子,包括TetR、ArgR2、CitR、RpiR、PurR等。进一步的EMSA、BLI等技术验证了这些调控因子与CLA合成基因启动子区的结合特性,并确定了具有结合能力的调控因子具体结合位点。通过构建调控因子敲除菌株和过表达菌株,结合CLA产量检测和转录水平分析,发现不同调控因子对CLA生物合成具有不同的调控作用。具体而言,TetR呈现正调控作用,能够促进CLA合成基因的表达;RpiR则起负调控作用,抑制CLA合成。ArgR2和CitR虽然未直接参与CLA的转录调控,但在调控因子间的相互作用中发挥了重要作用,显示出它们在CLA合成过程中起到间接调控的作用。此外,本研究通过RNA-seq高通量测序技术对植物乳杆菌AR195及其突变株的转录组进行了全面分析,揭示了CLA生物合成相关基因及其他调控因子表达的变化。基于研究结果,我们构建了CLA代谢调控因子间的协作网络,深入解析了CLA生物合成的转录调控机制。本研究为调控乳酸菌CLA生物合成的技术创新和产业应用奠定了重要基础。
