星形胶质细胞(As)水肿是脑缺血等众多疾病治疗面临的医学难题。依据渗透压是调控水进出细胞的驱动力,申请人实验室创建了角度中间纤维张力探针,将渗透压牵拉的骨架张力变化转换为实时光学信号,发现:缺血能刺激As微丝微管解聚,生成大量蛋白纳米颗粒,通过吸附阳离子,诱导As渗透势能上调及脑水肿发生,符合“唐南效应”。然而,“唐南效应”并未合理解释纳米颗粒吸附离子能力改变及游离阳离子减少对膜两侧电位的影响。本研究将引用纳米“双电层压缩”理论,解析脑缺血蛋白颗粒生成先诱导As低渗和超极化,激活HCN等电压门控离子通道,调控钠离子内流及电位恢复;但是后续T和L型钙通道导入的二价离子,反而促进蛋白颗粒释放一价阳离子,诱导阴离子内流、胞内高渗和水肿发生;同时揭示脑缺血胞内骨架解聚诱导蛋白颗粒生成的化学信号机制。藉此以降低胞内渗透势能为靶标,筛查治疗缺血性脑水肿的药物组合,探寻脑病治疗的新药研发方向。

Astrocyte edema is a medical problem in the treatment of many diseases such as cerebral ischemia. According to the preliminary research that osmotic pressure serves as the driving force regulating water flow across living cells, our laboratory created angle tension probes and integrated them into the intermediate filament, which converted the osmotic pressure variation in living cells into the optical signals. Our study found that ischemia stimuli resulted in the depolymerization of intracellular microfilament and microtubules, producing a mass of protein nanoparticles in astrocytes. The intracellular nanoparticles induced the up-regulation of permeability potential through the adsorption of cations, and then contributed to occurrence of cerebral edema. Although the above phenomenon is consistent with the "Donnan Effect" theory, it is insufficient to reasonably explain the variable ability of nanoparticles to adsorb ions and the effect of decreasing free cation on the membrane potential. In this present study, according to "the double-layer compression" theory, we will analyze that the generation of protein nanoparticles can induce hyperpolarization of astrocytes in response to ischemia, subsequently activate the various voltage-gated ion channels and upregulate intracellular ions, especially the T and L-type calcium channel. Influx of divalent ions enables to induce the release of monovalent ions from protein nanoparticles, resulting in up-regulation of osmotic pressure and occurrence of astrocyte edema. In addition, we will reveal that the molecular mechanisms underlying depolymerization of intracellular microfilament and microtubules and production of mass protein nanoparticles in cerebral ischemia. The study aims to reveal the pathogenesis of astrocyte swelling and brain edema in ischemia stimuli, develop new drugs in the area of osmotic potential, and explore the new pharmacological mechanisms and drug development direction for the treatment of brain edema.
神经细胞毒性水肿是脑缺血等众多疾病治疗面临的医学难题。依据渗透压是调控水进出细胞的驱动力,采用中间纤维张力探针将跨膜渗透压的力学效应转换为光学信号,更改了渗透压仪以颗粒总量评价细胞渗透压的机械检测方法,提出了与传统迥异的生物渗透压新理论。研究发现:.人体蛋白纳米颗粒(PN)能诱导膜电位电流的改变。抑制骨架解聚或炎症小体等PN生成,能缓解多种损伤因素刺激的神经细胞极化和水肿,也能拮抗血管紧张素刺激的肺内皮细胞超极化和肿胀发生;及下调高汞离子触发的膜电位持续去极化。而钙信号能调控PN诱导的去极化和超极化转换,这一现象符合双电层压缩理论。因而,PN对人体的主要作用是电调控,其作用类似于第二信使钙离子,两者协同水参与了膜电位调节,推测胞质游离蛋白含量差异能形成不同静息电位。.胞内外离子差异性通透对水的驱动,而不是离子总量,是调节生物渗透压力学效应的重要机制。等渗尿素能诱导神经细胞PN增加及胞内低渗萎缩破裂,而纯水低渗刺激能诱导神经细胞高渗膨胀和乳腺癌细胞低渗萎缩,提示细胞膨胀或萎缩与细胞内外离子数量无关。同时,钙激活电压依赖的TRPM4和TMEM16F等非选择性离子通道对水肿调节作用显著,提示钠、氯离子大量内流和钾离子少量通透对生物渗透压的调控。CaM和PKCα作为上述离子通道的增敏分子对生物渗透压调节也至关重要。而超极化激活的钾通道BK和IK及钾离子外排参与了尿素撤除诱导的神经细胞低渗萎缩调控。.质谱分析蛋白质组表明:胞质PN增多来源于多种蛋白的胞质转位,且受骨架蛋白和炎症小体等调节。.上述实验充分证明了生物渗透压是不同于传统渗透理论的新认识。基于此理论,我们识别蛋白错误折叠、炎症小体的生成以及骨架解聚调控了胞质总PN的含量及细胞力电协同活动,是人体脑水肿治疗的新药靶点。该研究更改了以“离子和水通道”为药物靶点治疗脑水肿的传统观点,为水肿靶向治疗提供新的思路。
