促性腺素抑制激素(GnIH)是禽类下丘脑关键生殖调控因子,能直接或间接通过GnRH抑制垂体促性腺激素分泌;然有研究提示,GnIH还能直接在性腺发挥调控作用,但对其机制知之甚少。项目研究马岗鹅:①卵泡发育过程中大白卵泡、小黄卵泡及大黄卵泡颗粒层和膜层中GnIH、GnIHR、FSHR、LHR、P450arom、StAR、INH、ACT表达变化;②光照调控繁殖季节性转换过程中,结合主动免疫GnIH,对产蛋、卵泡发育、以上基因在卵巢内表达及血液FSH、LH、PRL、P4、E2、INH水平的影响;③不同繁殖期(休产、产蛋、就巢),卵泡发育、以上基因在卵巢内的表达及血液FSH、LH、PRL、P4、E2、INH水平;④外源GnIH或抗GnIH抗体对颗粒细胞和膜细胞增殖、凋亡和以上基因表达及P4、E2、INH分泌的影响;揭示GnIH在卵巢内调控马岗鹅卵泡发育的分子机制,为完善禽类生殖调控理论提供科学依据。

As a key reproductive regulator in hypothalamus of avian, Gonadotropin inhibitory hormone (GnIH) can inhibit the gonadotropin secretion directly or indirectly through GnRH. However, previous studies have shown that GnIH could directly regulate follicular development in gonads, but little is known about its mechanism. Taking Magang goose as the object, this project would study: ①The gene expression patterns of GnIH、GnIHR、FSHR、LHR、P450arom、StAR、INH、ACT in the large white follicles (LWF), small yellow follicles(SYF) and the granulosa cells and theca layer of the large yellow follicles (LYF) during the follicular development. ②The egg laying, the follicular development, the change rule of the above genes in ovary and the levels of FSH, LH, PRL, P4, E2 and INH in blood during the seasonal transformation process of reproductive activity by artificial lighting, and the effects of active immunization with GnIH. ③The patterns of follicular development, expression of the above genes in the ovary and the blood levels of FSH, LH, PRL, P4, E2 and INH in different reproductive stages (no-breeding, laying and incubation). ④The effects of exogenous GnIH or anti-GnIH antibody on proliferation, apoptosis of granulosa cell and theca cell, and expression of the above genes and secretion of P4, E2 and INH. Those above results will reveal the molecular regulation mechanism of GnIH on follicular development in ovary of Magang goose, and provide scientific evidences for perfecting the reproductive control theory of birds.
促性腺素抑制激素(GnIH)是禽类下丘脑关键生殖调控因子。大量研究证实,GnIH在下丘脑-垂体层面能直接或间接通过GnRH抑制促性腺激素分泌来调控禽鸟类繁殖;但有研究提示,GnIH可能直接在性腺调控卵泡发育。项目以马岗鹅为对象,研究了GnIH在性腺直接调控卵泡发育的细胞分子机制,且在不同繁殖类型禽(鸟)类上进行比较研究(鸭、鹌鹑)。.结果显示,GnIH/GnIHR在卵巢颗粒细胞和膜细胞中均有表达,主要在颗粒细胞。外源GnIH能下调增殖相关基因表达,抑制颗粒细胞增殖,降低FSHR、LHR表达和类固醇生成;添加抗GnIH抗体或干扰GnIH均能明显促进颗粒细胞增殖,提高FSHR、LHR、PRLR表达,一定程度促进类固醇生成。在马岗鹅繁殖活动季节性转换过程中,随着繁殖活性退化,卵巢萎缩,下丘脑-垂体层面促生殖激素下降,VIP-PRL和GnIH/GnIHR表达上升,卵巢内GnIH/GnIHR表达上升,FSHR、LHR表达下降。主动免疫GnIH会明显抑制马岗鹅繁殖活动季节性退化,延长产蛋,促进卵巢内FSHR、LHR表达。在马岗鹅产蛋-就巢转换过程中,随着就巢发生,卵巢萎缩,卵泡发育减慢至停止,卵巢内GnIH/GnIHR、FSHR、LHR表达呈现一致变化,随着卵泡的发育而升高,类固醇生成逐步加强。在卵泡快速发展阶段,GnIH/GnIHR表达水平的增势减弱,FSHR和LHR表达增势则增强,LHR表达到排卵前达高峰。比较研究了光照对不同禽(鸟)类繁殖的影响及GnIH/GnIH在其中作用不同繁殖类型的禽(鸟)类,包括鹅(短日照季节性繁殖)、鸭(非季节性繁殖)、鹌鹑(长日照季节性繁殖)。揭示了GnIH/GnIH、VIP/PRL、OPN5+TSH-DIO2/DIO3在光照调控禽(鸟)类繁殖活动中发挥重要作用,发现OPN5+TSH-DIO2/DIO3在不同繁殖类型中介导机制不同,GnIH/GnIHR在性腺层面的调控机制基本相同,主要调控类固醇通路、促性腺激素受体的表达。.结果证实,GnIH/GnIHR直接在卵巢中表达,通过影响颗粒细胞和膜细胞增殖、凋亡及抑制FSHR、LHR表达和类固醇生成来调控卵泡发育。结果进一步揭示,GnIH在生殖轴下丘脑、垂体和性腺三个层面对繁殖活动发挥全面调控作用。
