英文摘要
Potato (Solanum tuberosum)is one of the most important food crops of the world,and the tuber shape directly impacts the appearance quality and economic value. Ro has been revealed to be a major locus controlling tuber shape. Previous studies had demonstrated that OVATE family protein 20 (StOFP20) was co-segregated with tuber shapes, and likely underlied the Ro locus. We obtained a population segregating for tuber shape, and fine-mapping delimited the causative locus to the same interval as that for Ro. Disabling of StOFP20 led to an altered tuber shape from round to oval shape, which proved the function of StOFP20 and lay a solid foundation for mechanism study. This project will : (1)utilize the methods of subcellular localization and in situ hybridization to reveal the subcellular localization and expression pattern of StOFP20; (2) understand the cellular difference in cell size, number or shape between round and oval tubers based on the histological analysis; (3) dissect the hormonal pathways that StOFP20 is involved on the basis of transcriptome analysis and hormonal response experiments; (4) screen and corroborate the interacting proteins with the yeast and plant experiment systems. The results will unravel the molecular mechanism of StOFP20 controlling tuber shape, which will provide molecular markers and new knowledge for developing elite potato varieties.
