Foot-and-mouth disease virus (FMDV), as an animal virus, seriously affects the healthy development of animal husbandry. Its endocytosis and specific mechanism has become a hot topic in virology research, which is of significance to the development of targeted vaccines for rapid prevention. We previously confirmed that FMDV can invade host cells by using the macropinocytosis pathway. By proteomics and gene interference, we further found that membrane protein CD44, as a cell adhesion molecule (CAMs) directly related to receptor tyrosine kinases (RTKs), is related to the early stage of FMDV infection. Therefore, this study will continue to trace whether CD44 proteins, as important molecules in the FMDV macropinocytosis pathway, are involved in initiating and/or regulating FMDV cellular endocytosis and what is specific mechanisms. The relationship between CD44 and RTKs activation in macropinocytosis, and the receptors and co-factors that may interact with CD44 proteins in macropinocytosis, and downstream kinase and GTPase signaling pathways of macropinocytosis in which receptors/co-factors together with CD44 jointly participate and play a regulatory mechanism on virus infection, were investigated at the cellular and in vivo levels, respectively. It not only provides the research data for the pathogenic mechanism of FMDV infection, but also provides the basis for the molecular design of effective targeted vaccines and the selection of the target sites of broad-spectrum antiviral agents.
