HBV大外膜蛋白preS1结构域的N端是识别受体的关键区域,我们曾利用这一区域的多肽发现了NTCP是HBV与HDV的功能性受体。此后,我们以及国内外其它团队在NTCP介导的病毒入侵机制方面取得了诸多进展。但是到目前为止,HBV脱包膜的机制还不清楚,preS1与NTCP是否在这一过程中发挥功能尚待研究。在原有工作基础上,我们分析发现紧邻受体结合位点的preS1中段含有一段疑似融合肽的区域,其疏水性对病毒感染性非常关键。结合以往研究,我们提出假说:preS1前段与NTCP结合后引起构象变化,激活preS1中段融合肽并引发HBV脱包膜的过程。我们将利用病毒反向遗传学、多肽功能鉴定、非天然氨基酸光交联、脂质体多肽展示等手段,深入探索preS1与NTCP介导HBV脱包膜的分子机制。阐明HBV脱包膜机制是全面理解HBV早期入侵过程的关键,对寻找新的药物靶点、开发新型抗HBV药物有重要意义。

The amino-terminal (N-terminal) region of preS1 domain on hepatitis B virus (HBV) large surface protein (L protein) is the key determinant for receptor binding. By using a photoactivatable preS1 lipopeptide, we identified sodium taurocholate cotransporting polypeptide (NTCP) as a functional receptor for both HBV and hepatitis δ virus (HDV). Later, we and others made significant progresses in studying the critical roles of NTCP in HBV entry. Yet, it remains enigmatic how HBV fusion was achieved and whether the preS1 domain and NTCP contribute to this process. Based on our extensive sequence analysis of the L protein and other preliminary data, we found that the preS1 middle sub-domain carries a conserved motif that resembles a viral fusion peptide, and its hydrophobicity is required for the viral infectivity. Here we hypothesize that: the N-terminal region of preS1 in the L protein interacts with NTCP and leads to a conformational change of preS1 to expose the potential fusion peptide in the preS1 middle sub-domain, which subsequently triggers the downstream membrane fusion and endosomal escape events. To test this hypothesis, we will employ viral reverse-genetics, peptide function analysis, unnatural amino acid based photo-crosslinking, and lipid-nanoparticle based peptide display to investigate the molecular mechanisms of HBV fusion. Understanding how HBV fuse with the endosomal membrane will provide a key piece of puzzle for the framework of HBV entry, and help identifying new drug targets and developing novel therapeutics.
我们曾鉴定出NTCP受体是乙肝病毒(HBV)及其卫星病毒丁肝病毒(HDV)入侵细胞的功能性受体,其中HBV L蛋白是负责受体识别的重要膜蛋白,其preS1结构域与NTCP的相互作用很可能在病毒早期融合过程中发挥重要作用,但其具体机制仍不清楚。本项目聚焦更囊膜病毒与受体相互作用以及膜融合的分子机制开展研究,综合分子生物学,反向遗传学,病毒学,生物化学等研究手段,探索HBV早期入侵过程中preS1与NTCP的相互作用如何影响下游入侵过程。实验结果提示preS1多肽的中段序列(49-73位氨基酸)对于HBV L外膜蛋白发挥功能至关重要,通过单点突变破坏序列疏水性或将其转化为跨膜域将使病毒失去感染能力;preS1多肽的中段序列需要在前段序列(2-48位氨基酸)与NTCP结合后发挥功能,且前段与中段序列的相对距离非常重要,这一相互作用与HBV成功完成脱包膜过程密切相关;另外,我们通过测试10种蝙蝠HBV病毒的preS1序列以及嵌合外膜蛋白,发现了两种蝙蝠HBV的preS1能够与人NTCP相互作用并介导入侵。在本项目支持下,我们在Viruses期刊发表了一篇关于利用preS1鉴定NTCP受体的综述论文,本项目负责人加入了国际乙肝基金会新兴学者、科学和医学顾问委员会。此外,基于本项目的相关研究思路与技术方法,本课题组还深入探索了冠状病毒与受体的相互作用以及膜融合的分子机制,提出了定制化病毒受体的新概念,在Nature (×2),Nat Commun, Cell Discovery等期刊以通讯作者身份发表多篇论文,获批国家优秀青年科学基金项目(2023年)。综上所述,本项目的实施为揭示HBV特殊的穿膜机制提供了重要线索,为理解病毒与受体相互作用以及早期入侵过程提供了重要依据。
