子宫颈腺癌发生率不断上升,预后差,严重影响妇女健康,故加强对子宫颈腺癌的基础研究,寻找潜在治疗靶点,是降低子宫颈腺癌死亡率的关键。本项目通过测序和验证发现新的环状RNA circSPIDR在子宫颈HPV相关腺癌组织中特异性表达下降,结合前期扎实研究基础,拟通过多种细胞功能检测明确circSPIDR对肿瘤细胞生长和侵袭的影响;利用多种细胞模型阐释HPV18 E7对circSPIDR的调控;利用FISH、RIP、荧光素酶报告基因检测、RNA-pulldown、蛋白质谱分析、CLIP-seq等多种检测手段解析circSPIDIR分别作为miR-431-5p海绵和与A1CF互作影响下游靶分子从而调控子宫颈HPV相关腺癌进展过程的分子机制;最后通过大样本组织验证和动物实验,证实circSPIDR的潜在临床应用价值和治疗价值。本项目为探索子宫颈HPV相关腺癌具有潜在临床应用价值的治疗靶点提供新的思路。

In China, the incidence of cervical adenocarcinoma is still increasing, threatening the health of women. However, our knowledge on cervical adenocarcinoma is limited. Strengthening the basic research on cervical adenocarcinoma and elucidating its molecular mechanism to identify therapeutic targets are the key steps to reducing the incidence of cervical adenocarcinoma. HPV-associated cervical adenocarcinoma is the main subtype of cervical adenocarcinoma. Our preliminary studies have identified a novel circular RNA named circSPIDR by circRNA sequencing, which was significantly downregulated in HPV-associated cervical adenocarcinoma tissues. On the basis of our preliminary reliable studies, we will explore the functional roles of circSPIDR in HPV-associated cervical adenocarcinoma cells. Combining with CRISPR targeted knockout and knock-in cell models, the HPV18 E7 regulatory mechanisom on circSPIDR will be explored to explain why circSPIDR was downregulated in HPV-associated cervical adenocarcinoma. Combining with FISH, RIP, Luciferase reporter assay, RNA-pulldown, Protein profiling, CLIP-seq and other detection methods, the regulatory molecular mechanisms of circSPIDIR acting as a miR-431-5p sponge and interact with A1CF to affect downstream moleculars will be investigated to improve the theory of circSPIDR function. Finally, large clinical tissue sample validation and animal models will be conducted to confirm the potential clinical application value and therapeutic value of the circSPIDR regulatory pathway. This project provides theoretical basis for elucidating the molecular mechanism of HPV-associated cervical adenocarcinoma, and also provides new ideas for exploring therapeutic targets with potential clinical value during the progression of HPV-associated cervical adenocarcinoma.
子宫颈癌严重危害妇女生命健康,目前子宫颈腺癌的发生率并无下降甚至上升,且子宫颈腺癌对放疗和化疗的敏感性不及鳞癌,预后更差。因此,寻找治疗子宫颈腺癌的分子靶标,提高子宫颈腺癌治愈率和降低死亡率已成为迫切需要解决的问题。本项目利用子宫颈正常组织和子宫颈腺癌组织进行全转录组测序,分析子宫颈腺癌circRNA、mRNA和miRNA的差异表达谱,发现并证实circSPIDR在子宫颈腺癌中特异性异常表达抑制。调控circSPIDR证明circSPIDR能够在体内外抑制子宫颈腺癌细胞恶性表型。进行同批组织的miRNA差异表达结果和circSPIDR结合位点特征比对,确定了miR-431-5p是circSPIDR结合的miRNA,两者存在结合位点,表达趋势符合ceRNA作用调控。确定了miR-431-5p对子宫颈腺癌细胞恶性表型的影响且miR-431-5p能够部分拮抗circSPIDR的作用。将同批组织的mRNA差异表达结果与miR-431-5p靶基因生信分析结果的对比分析,筛选出16个差异表达靶基因,验证实验显示circSPIDR仅调控CUBN和SORCS1的表达。通过一系列实验最终证实circSPIDR通过竞争性结合miR-431-5p解除后者对靶基因CUBN和SORCS1的抑制作用进而在宫颈腺癌细胞中发挥抑癌作用。通过RNA-pulldown实验及质谱分析,寻找到circSPIDR可能的结合蛋白。完成RNA-pulldown及Western Blot实验验证,证实circSPIDR与RPS3A的结合。实验证明circSPIDR通过与RPS3A相拮抗,调控DNA损伤修复过程以及调控细胞凋亡、细胞增殖等生物学表型。此外, circSPIDR能增加宫颈HPV相关腺癌对顺铂治疗的敏感性。体内动物实验证实circSPIDR具有协同顺铂抑制裸鼠移植瘤生长的作用且RPS3A能够拮抗circSPIDR与顺铂的协同肿瘤抑制作用。最后,在大样本组织标本中分析了circSPIDR对于宫颈腺癌患者的临床价值。我们的研究阐明了circSPIDR在调控子宫颈腺癌进展中的作用及相关分子机制,为寻找抑制宫颈腺癌进展的有效治疗靶点提供充分的科学依据。本项目按要求完成,发表高质量SCI论文6篇,已接收1篇,申请国家发明专利1项,获得省部级科技奖1项,参加国内外学术交流多次,培养博士研究生2名,硕士研究生2名。
