The surface distribution of antigenic sites on ejaculated and epidid ymal rabbit spermatozoa was investigated before and after incubation in media presumed capacitative of spermatozoa. It was found that certain components overlying the acrosomal region were effectively removed by both isotonic and hypertonic media resulting in "bare" patches in the hemocyanin-labeled antibody region. Completely unlabeled acrosomes were often observed in cells treated with hypertonic medium whereas the postnuclear sheath exhibited numerous labeled sites. Freeze-etching revealed an extensive surface coat layer over the sperm head which was considerably less evident over the sperm tail. There was an apparent correlation between this distribution and the pattern of the antibody label. The results are discussed in relation to the possible changes in sperm head membrances prior to the acrosomal reaction.
