The hypothesis of heterogeneity of 5-HT (5-hydroxytryptamine, serotonin) receptors is supported by a wealth of pharmacological, electrophysiological and biochemical data. The 5-HT receptors have been classified into three main types, termed 5-HTl-like, 5-HT 2 and 5-HT 3 (Bradley et al., 1986). While both 5-HTl-like and 5-HT 2 receptors have long been evidenced in the CNS and in the periphery, 5-HT 3 binding sites have been found only recently in the brain (Kilpatrick et al., 1987). We now report on the localisation of 5-HT 3 recognition sites in the mouse CNS by means of the potent and selective 5-HT 3 antagonist [3H] ICS 205-930, recently used to label 5-HT 3 sites on NG108-15 neuroblastoma-glioma cells (Hoyer and Neijt, 1987). Mice (male, OF1, 22-27 g) were killed by decapitation and the brains were quickly removed and frozen on dry ice. Sections 10# m thick were cut with a microtome cryostat and thaw-mounted onto gelatin-coated slides. The sections were preincubated for 30 min at room temperature in 170 mM Tris. HC1 buffer containing 150 mM NaCI, 4 mM CaC12 and 0.01% ascorbic acid. 5-HT 3 recognition sites were then labelled by incubating the slides for 60 min in the same medium (room temperature) but containing 1.5 nM [3H] ICS 205-930 (33 Ci/mmol).
