METHODSExperimental procedures. Frog sartorii, strips of frog ventricle, or rabbit psoas muscles were tied to wooden sticks at rest length and placed in a 1: 1 glycerol water mixture at 0 C for 2 days. Thereafter, the solution was exchanged, and the muscles were stored in a freezer at-15 C until used. In some experiments hard-boiled chicken egg white was used. This was selected as a protein which is isoelectric at pH 4.6 (5) and so has negative charges at neutral pH. The protein was boiled to simplify handling; it was assumed that the heat coagulation did not cause a large shift in the isoelectric point.Potential measurements were made with KCl-filled glass microelectrodes. The electrodes were inserted into the tissue mass with a micromanipulator, just as a cell is impaled. The potentials found are sensitive to pressure and occlusion of the tip. To minimize these effects, the electrode resistance was monitored, and the potential changes found on “impaling” the glycerinated muscles or the egg white were accepted only
