CD8 is a cell surface glycoprotein expressed by most thymocytes and by mature T lymphocytes that recognize class I major histocompatibility complex (MHC) molecules. Most CD8 cells are cytotoxic or suppressor in function. It has been postulated that the function of this molecule is to increase the avidity of the interaction between T cells and target cells or antigen-presenting Recent evidence provides strong support for this notion. 4d The presumed ligand for CD8 is a nonpolymorphic region of class I MHC molecule^,^-^ but there has been no direct demonstration of such binding to date. To explain data in which monoclonal antibodies (mAbs) specific for CD8 inhibit T-cell activation induced by lectins or anti-CD3 mAbs, it has alternatively been suggested that the function of CD8 might be to deliver a negative signal to the T ell.^.^ However, there are currently no data to indicate transmission of such a signal. In this report I will review some of our recent studies of the molecular structure, expression, and function of CD8.
